Connections between the mechanical properties of DNA and biological functions have been speculative due to the lack of methods to measure or predict DNA mechanics at scale. Recently, a proxy for DNA mechanics, cyclizability, was measured by loop-seq and enabled genome-scale investigation of DNA mechanics. Here, we use this dataset to build a computational model predicting bias-corrected intrinsic cyclizability, with near-perfect accuracy, solely based on DNA sequence.

Efficient prime editor (PE) delivery in vivo is critical for realizing its full potential in disease modeling and therapeutic correction. Although PE has been divided into two halves and delivered using dual adeno-associated viruses (AAVs), the editing efficiency at different gene loci varies among split sites. Furthermore, efficient split sites within Cas9 nickase (Cas9n) are limited.

The Constrained Disorder Principle (CDP) defines all systems in nature by their degree of inherent variability. Per the CDP, the intrinsic variability is mandatory for their proper function and is dynamically changed based on pressures. The CDP defines the boundaries of inherent variability as a mechanism for continuous adaptation to internal and external perturbations, enabling survival and function under dynamic conditions.
