The ability of a phosphine-appended-2,2'-bipyridine ligand ((Ph2P)2bpy) to serve as a platform for late-stage ligand modifications was evaluated using tetrahedral (Ph2P)2bpyFeCl2. We employed a post-metalation Staudinger reaction to install a series of functionalized arenes, including those containing Brønsted and Lewis acidic groups. This reaction sequence represents a versatile strategy to both tune the ligand donor properties as well as directly incorporate appended functionality.

Optically pure 1,2-diols and 1,3-diols are the most privileged structural motifs, widely present in natural products, pharmaceuticals and chiral auxiliaries or ligands. However, their synthesis relies on the use of toxic or expensive metal catalysts or suffer from low regioselectivity. Catalytic asymmetric synthesis of optically pure 1,n-diols from bulk chemicals in a highly stereoselective and atom-economical manner remains a formidable challenge.

A ligand-promoted oxidative dehydrogenation C-H alkenylation of indoles and olefins was achieved using commercial and low-cost Co(NO)·6HO as a catalyst and Mn(OAc) as an oxidant. The design and selection of electrically unique methyl-substituted salicylaldehyde as a ligand is the key to achieve this transformation. This protocol can introduce an indole backbone into diverse bioactive molecules such as ibuprofen, naproxen, and Estrol for late-stage synthetic modification, which has potential applications in the discovery of drug molecules containing an indole motif.

A copper-catalyzed regioselective annulation reaction, conducted without ligands or oxidants, has been developed for the preparation of multisubstituted furans from the readily available starting materials, β-keto esters and propargyl acetates. This process accommodates a wide range of functional groups, resulting in furan skeletons with diverse substitution patterns. The method's potential synthetic utility is highlighted by its applicability in gram-scale preparations and late-stage modifications of natural products.

Genetically encoded libraries (GEL) are increasingly being used for the discovery of ligands for "undruggable" targets that cannot be addressed with small molecules. Foundational GEL platforms like phage-, yeast-, ribosome-, and mRNA-display have enabled the display of libraries composed of 20 natural amino acids (20AA). Unnatural amino acids (UAA) and chemical post-translational modification (cPTM) expanded GEL beyond the 20AA space to yield unnatural linear, cyclic, and bicyclic peptides.
