Sema4D, a member of the immune semaphorin family, plays crucial roles in the immune regulation, bone resorption and nervous system. It is also involved in angiogenesis and tumour progression. However, systemic studies on the correlation between Sema4D expression and the immune infiltration or clinical outcomes in tumours are still limited. Here, we analysed the landscape of Sema4D expression and its prognostic value in the cancer genome atlas pan-cancer as well as the correlation between Sema4D and immune cell infiltration by Tumour Immune Estimation Resource and Gene Expression Profiling interactive analysis online tools. Results showed that a higher Sema4D expression was significantly correlated with a favourable overall survival in diverse solid tumours including bladder cancer (Hazards Ratio (HR)=0.68, = .0095), kidney renal clear cell carcinoma (HR = 0.61, = .0016), melanoma (HR = 0.58, = 6.6e-05) and thymoma (HR = 0.1, = .011). Interestingly, Sema4D expression has positive correlation with various tumour infiltrating immune cells and immune cell biomarkers in these tumours. These results suggest that Sema4D could be a prospective biomarker for calculating hazard ratio of tumour patients and their tumour immune infiltration levels.

Distal metastases result from metastatic microenvironment and tumour epithelial cell interactions, the cellular heterogeneity of primary colorectal cancer (CRC) and liver metastases (LM) was evaluated by integrating single-cell sequencing data, and the collected gene expression data from metastatic epithelial cell subsets was used to construct a prognostic model and to identify intercellular receptor-ligand interactions between epithelial and immune cells in CRC and LM. Multiplex immunofluorescence staining, and in vitro wound healing, cell migration and cell apoptosis assays were performed to further explore the biological relevance of identified potential regulatory molecules. In this study, approximately 17 epithelial cell subtypes were detected, with Epi-11 cells being highly expressed in LM tissues compared with CRC samples.

The crosstalk between microglia inflamed in multiple sclerosis (MIMS) and astrocytes inflamed in MS (AIMS) is a crucial factor in the formation of the central inflammatory microenvironment and neurotoxicity. Astragalus polysaccharides (APS), an important bioactive component extracted from the dried root of Astragalus, was previously found by our team to attenuate the formation of pro-inflammatory microglia and neurological dysfunction in the experimental autoimmune encephalomyelitis (EAE) mice, a classic model of MS. To investigate the effect of APS on the MIMS-AIMS crosstalk and its underlying mechanism, in this study, a mouse model of EAE and a co-culture model of microglia-astrocytes in vitro were established.
