AI Article Synopsis

  • Ameloblastoma is a benign yet locally aggressive tumor that arises from the odontogenic apparatus in the jaws, showing varied clinical and biological behaviors, particularly between its follicular and plexiform types.
  • The study aimed to investigate the expression of the NF-κB protein in these two histological types of ameloblastomas, using immunohistochemical techniques to compare their expression patterns.
  • Results indicated that NF-κB p65 was more frequently expressed in plexiform ameloblastomas, particularly in the outer preameloblast-like cells, suggesting a potential biological mechanism that differs between the two types.

Article Abstract

Background: Ameloblastoma among benign tumors holds a unique position by its locally destructive and invasive nature. Tumors that originate from the odontogenic apparatus or its remnants in the jaws show diverse clinical presentations, behavior and histologic patterns. The differed biological behavior behind follicular and plexiform ameloblastomas has never attained completeness because of the lack of rhythmic correlation regarding the exact mechanism. Nuclear factor-kappa B (NF-κB) pathways play a crucial role in survival, death and differentiation during physiologic and pathologic conditions. With this background, the study has been aimed to investigate the expression of NF-κB in follicular and plexiform ameloblastomas.

Objective: The objective of this study was to analyze the immunohistochemical expression pattern of NF-κB in ameloblastoma and to compare the immunohistochemical expression pattern of NF-κB among the histological types of ameloblastoma, follicular and plexiform patterns.

Methodology: Total 20 ameloblastomas (10 follicular, 10 plexiform) were immunostained with antihuman NF-κB p65 mouse IgG monoclonal antibody, and the pattern of staining is statistically analyzed using Chi-square test with the level of significance ( < 0.05).

Results: Twelve (3 follicular, 9 plexiform) out of 20 ameloblastomas showed immunoreactivity to NF-κB p65. In ameloblastoma, only the peripheral preameloblast-like tall columnar cells showed reactivity, whereas the stellate reticulum-like cells are immunonegative. The staining pattern was membranous in the immunoreactive cells. The results were studied with the associated and inducing pathways from the literature, and a possible mechanism has been proposed.

Conclusion: The expression pattern of NF-κB was found to be higher in plexiform ameloblastoma than follicular ameloblastoma.

  • - The article examines a rare case of ameloblastoma found in a 74-year-old man, impacting the right maxilla, maxillary sinus, and nasal cavity, initially misdiagnosed as squamous cell carcinoma.
  • - A biopsy revealed uncertain origins for the tumor's multiple histologic patterns, leading to a confirmed diagnosis after surgical resection, highlighting challenges in clinical classification based on WHO's 2022 guidelines.
  • - The literature review of 45 similar cases showed that maxillary ameloblastoma often invades crucial structures, with common extensions to the nasal cavity and orbit, and emphasizes the difficulty in differentiating between primary sinonasal and gnathic origins.

Ameloblastoma of the Maxilla With Distant Metastases to the Lungs: A Case Report and Literature Review.

Maxillary ameloblastoma is one of the rarest odontogenic epithelial tumors encountered, as 80% of ameloblastomas are seen within the mandible. Ameloblastoma is usually incidentally detected in the third to fourth decades of life, as most patients remain asymptomatic; yet some patients may complain of a slowly growing, painless swelling. We present a case of maxillary ameloblastoma with pulmonary metastasis along with a brief literature review.

Assistant Professor of Oral Pathology, Oral Pathology Department, Faculty of Dentistry, Tanta University, Egypt.

Background: The current study aims at investigating gap junctions which allow cells to connect with one another. Such process is essential for cell differentiation and the preservation of diverse cell functions. It is noticeable that connexin 43 (Cnx43) was differentially expressed in ameloblasts and odontoblasts in the processes of odontogenesis.

Background: Various stemness markers (SOX2, OCT4, and NANOG) have been studied in odontogenic cysts and tumors. However, studies on SALL4 having similar properties of stemness has not been documented. Additionally, insight into fascin as a migratory molecule is less explored.

Department of Oral and Maxillofacial Surgery, Massachusetts General Hospital, Boston, MA, USA. Electronic address:

Ameloblastoma, odontogenic keratocyst (OKC), and dentigerous cyst (DC) can have similar radiographic and histological appearances. The purpose of this study was to determine the utility of BRAF immunohistochemistry in discerning mandibular ameloblastomas from OKCs and DCs. This retrospective cohort study included patients treated between 1998 and 2018.
