Alkyl hydroxycinnamates (AHs) is a group of molecules of biotechnological interest due to their cosmetic, food, and pharmaceutical applications. Among their most interesting uses are as UV protectants, skin depigmentation agents, and antioxidant ingredients which are often claimed for their antitumoral potential. Nowadays, many sustainable enzymatic approaches using low-cost starting materials are available and interesting immobilization techniques are helping to increase the reuse of the biocatalysts, allowing the intensification of the processes and increasing AHs accessibility. Here a convenient summary of AHs most interesting biological activities and possible applications is presented. A deeper analysis of the art state to obtain AHs, focusing on most employed enzymatic synthesis approaches, their sustainability, acyl donors relevance, and most interesting enzyme immobilization strategies is provided.Key points• Most interesting alkyl hydroxycinnamates applications are summarized.• Enzymatic approaches to obtain alkyl hydroxycinnamates are critically discussed.• Outlook of enzyme immobilization strategies to attain alkyl hydroxycinnamates.

Article Synopsis
  • Golden berries (Physalis peruviana) are recognized for their neuroprotective qualities and traditional medicinal uses, prompting a study to assess their effectiveness against neurotoxicity caused by cyclophosphamide (CP).
  • The study utilized a combination of advanced chromatography and mass spectrometry to analyze the chemical composition of golden berries, alongside behavioral tests and histological evaluations in an animal model to gauge the neuroprotective effects of total leaf extract (TLE).
  • Results indicated that TLE significantly preserved hippocampal neurons and enhanced cognitive performance by reducing acetylcholinesterase activity and oxidative stress, while also restoring redox balance through the inhibition of inflammatory pathways.

Amino acid-based poly(ester urea)s are an attractive class of polymers that are of interest for a variety of biomedical applications. Generally, amino acid-based poly(ester urea)s are prepared by polymerization of diamines, which are obtained from the corresponding amino acids and aliphatic diols. This article presents an alternative synthetic strategy that uses diamine monomers obtained from aromatic, 4-hydroxycinnamic acid-derived diols.

Our research group previously identified graviquinone (1) as a promising antitumor metabolite that is formed in situ when the antioxidant methyl caffeate scavenges free radicals. Furthermore, it exerted a DNA damaging effect on cancer cells and a DNA protective effect on normal keratinocytes. To expand and explore chemical space around qraviquinone, in the current work we synthesized 9 new alkyl-substituted derivatives and tested their in vitro antitumor potential.

Fluorescent Labeling of a Target Protein with an Alkyl Diazirine Photocrosslinker Bearing a Cinnamate Moiety.

Chem Asian J

June 2024

Laboratory of Biorecognition Chemistry, Faculty of Pharmaceutical Sciences, Academic Assembly, University of Toyama, 2630 Sugitani, Toyama, 930-0194, Japan.

A novel fluorogenic alkyl diazirine photocrosslinker bearing an o-hydroxycinnamate moiety has been developed for identification of the targets of bioactive molecules. The o-hydroxycinnamate moiety can be converted to the corresponding 7-hydroxycoumarin derivative, which should be created on the interacting site within the photocaptured target protein. The label yield and fluorescence intensity have been immensely improved in comparison with our previous aromatic crosslinkers to facilitate target identification in small quantities.

ANCUT1, a novel thermoalkaline cutinase from Aspergillus nidulans and its application on hydroxycinnamic acids lipophilization.

Biotechnol Lett

June 2024

Departamento de Alimentos y Biotecnología, Facultad de Química, Universidad Nacional Autónoma de México (UNAM), Ciudad Universitaria, CP 04510, Ciudad de México, Mexico.

One of the four cutinases encoded in the Aspergillus nidulans genome, ANCUT1, is described here. Culture conditions were evaluated, and it was found that this enzyme is produced only when cutin is present in the culture medium, unlike the previously described ANCUT2, with which it shares 62% amino acid identity. The differences between them include the fact that ANCUT1 is a smaller enzyme, with experimental molecular weight and pI values of 22 kDa and 6, respectively.
