This study investigated the expression of components of the renin-angiotensin system (RAS) by cancer stem cells (CSCs) we have recently demonstrated in renal clear cell carcinoma (RCCC). Fifteen RCCC tissue samples underwent immunohistochemical staining for components of the RAS: renin, pro-renin receptor (PRR), angiotensin-converting enzyme (ACE), angiotensin-converting enzyme 2 (ACE2), and angiotensin II receptor 2 (ATR). Immunofluorescence co-staining or double immunohistochemical staining of these components of the RAS with stemness-associated markers OCT4 or KLF4 was performed on two of the samples. Protein and transcript expression of these components of the RAS in six RCCC tissue samples was investigated using western blotting and reverse transcription quantitative polymerase chain reaction (RT-qPCR), respectively. In addition, angiotensin II receptor 1 (ATR) was investigated using RT-qPCR only. Immunohistochemical staining demonstrated expression of renin, PRR, and ACE2 in 11, 13, and 13 out of 15 RCCC samples, respectively, while ATR was expressed in all 15 samples. ACE was detected in the endothelium of normal vasculature only. Double immunohistochemical staining demonstrated localization of ACE2, but not renin, to the KLF4+ CSCs. Immunofluorescence staining showed localization of PRR and ATR to the OCT4+ CSCs. Western blotting confirmed protein expression of all components of the RAS except renin. RT-qPCR demonstrated transcript expression of all components of the RAS including ATR, but not ATR, in all six RCCC tissue samples. This study demonstrated expression of PRR, ACE2, and ATR by the CSCs within RCCC. Further studies may lead to novel therapeutic targeting of CSCs by manipulation of the RAS in the treatment of this aggressive cancer.

The chloroplast RNA-binding protein CP29A supports expression during cold acclimation.

Molecular Genetics, Institute of Biology, Faculty of Life Sciences, Humboldt Universität zu Berlin, Berlin 10115, Germany.

The chloroplast genome encodes key components of the photosynthetic light reaction machinery as well as the large subunit of the enzyme central for carbon fixation, Ribulose-1,5-bisphosphat-carboxylase/-oxygenase (RuBisCo). Its expression is predominantly regulated posttranscriptionally, with nuclear-encoded RNA-binding proteins (RBPs) playing a key role. Mutants of chloroplast gene expression factors often exhibit impaired chloroplast biogenesis, especially in cold conditions.

Orientation selectivity properties for the affine Gaussian derivative and the affine Gabor models for visual receptive fields.

J Comput Neurosci

Computational Brain Science Lab, Division of Computational Science and Technology, KTH Royal Institute of Technology, SE-100 44, Stockholm, Sweden.

This paper presents an in-depth theoretical analysis of the orientation selectivity properties of simple cells and complex cells, that can be well modelled by the generalized Gaussian derivative model for visual receptive fields, with the purely spatial component of the receptive fields determined by oriented affine Gaussian derivatives for different orders of spatial differentiation. A detailed mathematical analysis is presented for the three different cases of either: (i) purely spatial receptive fields, (ii) space-time separable spatio-temporal receptive fields and (iii) velocity-adapted spatio-temporal receptive fields. Closed-form theoretical expressions for the orientation selectivity curves for idealized models of simple and complex cells are derived for all these main cases, and it is shown that the orientation selectivity of the receptive fields becomes more narrow, as a scale parameter ratio , defined as the ratio between the scale parameters in the directions perpendicular to vs.

LPCAT1, the Enzyme Responsible for Converting LPC to PC, Promotes OPC Differentiation In Vitro.

Myelin is the key structure for high-speed information transmission and is formed by oligodendrocytes (OLs) which are differentiated from oligodendrocyte precursor cells (OPCs) in the central nervous system. Lipid is the main component of myelin and the role of lipid metabolism-related molecules in myelination attach increasing attention. Lysophosphatidylcholine acyltransferase 1 (LPCAT1) mediates the conversion of lysophosphatidylcholine (LPC) to phosphatidylcholine (PC), and its role in myelination draws our interest as LPC is a classical demyelination inducer and PC is a major component of myelin.

Liver cancer remains a significant global health challenge, characterized by high incidence and mortality rates. Despite advancements in medical treatments, the prognosis for liver cancer patients remains poor, highlighting the urgent need for novel therapeutic approaches. Traditional Chinese medicine (TCM), particularly (CB), has shown promise in addressing this need due to its multi-target therapeutic mechanisms.
