We have synthesized new magnetic resonance imaging (MRI) T contrast agents (CA1 and CA2) that permit the activatable recognition of the cellular vicinal thiol motifs of the protein thioredoxin. The contrast agents showed MR relaxivities typical of gadolinium complexes with a single water molecule coordinated to a Gd center (i.e., ~4.54 mMs) for both CA1 and CA2 at 60 MHz. The contrast agent CA1 showed a ~140% relaxivity enhancement in the presence of thioredoxin, a finding attributed to a reduction in the flexibility of the molecule after binding to thioredoxin. Support for this rationale, as opposed to one based on preferential binding, came from H-N-HSQC NMR spectral studies; these revealed that the binding affinities toward thioredoxin were almost the same for both CA1 and CA2. In the case of CA1, T-weighted phantom images of cancer cells (MCF-7, A549) could be generated based on the expression of thioredoxin. We further confirmed thioredoxin expression-dependent changes in the T-weighted contrast via knockdown of the expression of the thioredoxin using siRNA-transfected MCF-7 cells. The nontoxic nature of CA1, coupled with its relaxivity features, leads us to suggest that it constitutes a first-in-class MRI T contrast agent that allows for the facile and noninvasive monitoring of vicinal thiol protein motif expression in live cells.

Objective: This study explored the value of stomach ultrasound reporting and data system (Su-RADS) and two-dimensional shear wave elastography (2D-SWE) in the diagnosis of benign and malignant lesions of the gastric wall, evaluating the feasibility of combining the two methods for the diagnosis of gastric wall lesions.
Methods: 113 patients with gastric wall lesions were examined after oral gastric ultrasound contrast agent, and the grades of the gastric wall lesions were classified according to Su-RADS. Moreover, 2D-SWE was performed to measure the E value of the lesions.

A dual-signal aptamer-based assay utilizing colorimetric and fluorescence techniques was developed for the determination of zearalenone (ZEN). The CdTe quantum dots, serving as the fluorescent signal source, were surface-modified onto FeO@SiO and subsequently functionalized with the aptamer. The COF-Au was modified with complementary chain, which possessed peroxide (POD)-like enzyme properties, and could catalyze the peroxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to ox TMB, resulting in the generation of colorimetric signals.

Biliary and pancreatic tract stenosis are hallmark symptoms in pancreaticobiliary diseases, transcending malignancy. Endoscopic techniques are pivotal for biliary/pancreatic drainage; however, challenging scenarios arise when attempting to pass a guidewire (GW) through obstruction. Cholangioscopy-assisted GW placement has proven valuable, but challenges persist in its execution, particularly in maneuvering the GW through cholangioscopy.

Objective: Abnormal mechanical stress is intimately coupled with osteoarthritis (OA). Microtubules play a vital role in the regulation of mechanotransduction and intracellular transport. The purpose of the present study was to investigate the impact of stress-induced microtubule impairment on intracellular transport and lipid droplet (LD) accumulation in chondrocytes.

Background: Usefulness of hysteroscopic metroplasty to improve reproductive outcomes is controversial and debated among reproductive specialists and, consequently, patients.
Methods: We performed a cross-sectional analysis to assess the quality, reliability, and level of misinformation in YouTube, Instagram, and TikTok videos about hysteroscopic metroplasty. Videos on each social network retrieved using "hysteroscopy" and "septate uterus" or "uterine septum" as keywords were assessed using Patient Education Materials Assessment Tool for audio-visual (PEMAT A/V) content, the modified DISCERN (mDISCERN), Global Quality Scale (GQS), Video Information and Quality Index (VIQI) and Misinformation assessment.
