Pleurospermum (Apiaceae) species possess a wide range of biological properties viz. analgesic, anti-inflammatory, antimalarial, and so on. Pleurospermum candollei (DC.) Benth. Ex C. B. Clark. is reported to cure diarrhea, gastric, respiratory, stomach, abdominal, joint, and back pain problems. In addition, it is also used for both male and female infertility. The present study deals with an efficient technique using high-speed countercurrent chromatography for separation of chemical components from the methanol extract of P. candollei. Notably, nine main compounds namely luteolin 7-O-glucoside (1), oxypeucedanin hydrate (2), pabulenol (3), bergapten (4), heptadecanoic acid (5), (E)-isoelemicin (6), trans-asarone (7), α-linolenic acid (8), and isoimperatorin (9) were very efficiently separated and isolated in pure form. Multiple injections were applied followed by two off-line recycling high-speed countercurrent chromatography. The inhibitory effect of nitric oxide production of all compounds was tested in the presence of 200 ng/mL lipopolysaccharide in RAW264.7 mice macrophage cells. The results demonstrated that compounds 7 and 8 effectively inhibited nitric oxide production, with IC values of 28.44 and 53.18 μM, respectively. This study thus validates the traditional claim of using P. candollei. Taken together, these findings will be useful in future research to find a potential candidate with anti-inflammatory properties.

is a lesser-known plant species belonging to the genus that grows in Kazakhstan. The aim of this study was to characterize the composition of the ethanolic, water, and hydroethanolic extracts from the aerial parts of by HPLC-ESI-QTOF-MS/MS to isolate the major compound isoquercitrin by HSCCC (High-Speed Counter-Current Chromatography) and to determine the cytotoxicity and anti-inflammatory potential of the extracts produced with this plant. Fingerprinting of the analyzed extracts showed the presence of a multitude of metabolites comprising polyphenols, organic acids, and coumarins, and only trace quantities of cardiac glycosides in the analyzed samples.

Natural products play a vital role in human health because people find that they can reduce the incidence of many diseases, such as cancer and cardiovascular diseases. Therefore, researchers have developed many effective techniques for screening and separating active compounds from natural products. High-speed countercurrent chromatography is a unique unsupported liquid-liquid chromatography technology that has been widely used in the separation of natural products.

In this work, different injection modes (single injection, overlapping injection, and continuous injection) of countercurrent chromatography were used and compared for the isolation and purification of glabridin from the crude extract of Glycyrrhiza glabra. The two-phase solvent system consisting of n-hexane/ethyl acetate/methanol/water (5:4:5:4, v/v) was employed as stationary and mobile phases. Compared with single injection mode, both overlapping injection and continuous injection modes exhibited higher separation efficiency at the same sample loading capacity.

October 2024

Laboratório de Produtos Naturais (TecBio), Farmanguinhos, FIOCRUZ Foundation, Rua Sizenando Nabuco 100, Rio de Janeiro 21041-250, RJ, Brazil.

Article Synopsis
  • The text discusses the isolation of two cytotoxic compounds, aurelianolides A and B, from a plant in the Solanaceae family, which shows potential as anticancer drugs.
  • Traditional methods for obtaining these compounds are costly and time-consuming, prompting a study to explore a more efficient technique using high-speed countercurrent chromatography (HSCCC).
  • The study achieved a successful separation of the withanolides in a single step, resulting in high-purity yields of aurelianolides A and B, significantly improving the process compared to classical methods.

Article Synopsis
  • The study used Novozyme 435 lipase to perform a biocatalytic transesterification reaction on esculin, aimed at increasing its hydrophobicity and improving solubility for commercial use.
  • The reaction occurred at 60 °C and produced two main products: a monoacylated and a diacylated form of esculin, with the diacylated form being the dominant product at a conversion rate of 53.55%.
  • Both products were purified using high-speed counter-current chromatography and characterized by NMR, demonstrating a fast and efficient method for enhancing the practical applications of esculin through the creation of acetylated derivatives.
