White light phase-shifting interference microscopy (WL-PSIM) is a prominent technique for high-resolution quantitative phase imaging (QPI) of industrial and biological specimens. However, multiple interferograms with accurate phase-shifts are essentially required in WL-PSIM for measuring the accurate phase of the object. Here, we present single-shot phase-shifting interferometric techniques for accurate phase measurement using filtered white light (520±36 nm) phase-shifting interference microscopy (F-WL-PSIM) and deep neural network (DNN). The methods are incorporated by training the DNN to generate (a) four phase-shifted frames and (b) direct phase from a single interferogram. The training of network is performed on two different samples i.e., optical waveguide and MG63 osteosarcoma cells. Further, performance of F-WL-PSIM+DNN framework is validated by comparing the phase map extracted from network generated and experimentally recorded interferograms. The current approach can further strengthen QPI techniques for high-resolution phase recovery using a single frame for different biomedical applications.

Not out of the woods: perspectives from patients with triple-negative breast cancer.

Support Care Cancer

December 2024

Massachusetts General Hospital, Yawkey Center, Suite 9A, 55 Fruit St, Boston, MA, 02114, USA.

Purpose: Patients with triple-negative breast cancer (TNBC) are at high risk for breast cancer recurrence and metastatic disease, yet the scholarly literature on the distress and uncertainty of this vulnerable population is limited. This study aimed to characterize the experiences of patients with TNBC and obtain feedback about the development of a supportive care intervention targeted to this population's psychosocial needs.

Methods: From 9/2021 to 2/2023, we purposefully recruited 23 patients with stage I-III TNBC who recently completed curative therapy and conducted a parallel mixed qualitative and quantitative study.

In this study, we present lateral scanning white light interferometry (LS-WLI), where phase-shifting algorithms are applied to inspect the topography of a large field of view (FOV) with high-speed measurements. At a point, the interference signal must be acquired with a specific condition to adapt the phase-shifting algorithm. This means that all points have two points, of which the phase difference is π/2, when the number of points acquired in a phase period is multiple of 4, despite increasing the data points in a period.

Fresnel incoherent correlation holography (FINCH) records coaxial holograms for wide-field 3D imaging with incoherent light, but its temporal phase-shifting strategy makes dynamic imaging challenging. Here, we present a compact, portable single-shot mirrored phase-shifting (SSPMS) module that can be easily integrated into the FINCH system, achieving secondary modulation of self-interference beams to enable the simultaneous acquisition of four phase-shift holograms in a single exposure. Compared with previously reported methods that use diffraction gratings to spatially separate self-interference beams at specific angles, this module duplicates a laterally shifted mirrored beam using a simply modified Michelson interferometer, so the phase-shifting holograms obtained via this module are free from optical aberrations or higher-order diffracted light noises.

Article Synopsis
  • Quantitative phase microscopies (QPMs) enhance bio-imaging by providing critical data on mass distribution and transport, which is not achievable through fluorescence techniques and are label-free, avoiding issues like photobleaching.!*
  • The review compares eight QPM techniques, including digital holographic microscopy and phase-shifting interferometry, focusing on their accuracy and measurement capabilities using a custom-developed numerical toolbox for simulations.!*
  • Results indicate that DHM and PSI are robust against artefacts but can be affected by coherent noise, while other techniques show a balance between measurement precision and accuracy, with some experiencing limitations due to inherent artefacts, especially with larger samples like eukaryotic cells.!*

Dynamic deformation measurement with 2-frame phase-shifting speckle interferometry based on speckle statistics and wavefront multiplexing.

Phase-shifting speckle interferometry could achieve full-field deformation measurement of rough surfaces. To meet the dynamic requirement and further improve the accuracy, a two-step synchronous phase-shifting measurement system is established based on the polarization-sensitive phase modulation ability of a liquid crystal spatial light modulator; by multiplexing the reference wavefront, an accurate phase shift is generated between two independent recording channels, and a common-path self-reference vortex interference structure is built for precise spatial registration. Meanwhile, according to the speckle statistical principle, a novel two-frame phase-shifting algorithm as well as a two-step spatial registration strategy is presented to strengthen the robustness of intensity and position differences caused by spatial-multiplexing; thereby, accurate transient deformation can be directly obtained from phase-shifting speckle interferograms recorded before and after deformation.
