Aim: This study focuses on the design and investigation a transethosomal formulation for enhanced topical delivery and improved analgesic activity of caryophyllene oxide. In addition, this work explores new potential mechanisms of analgesic activity of the active compound including alpha-amino-3-hydroxy-5-methyl-4-isooxazole-propionic acid (AMPA) and Cyclooxygenase 2 (COX-2).
Methods: The transethosomal system containing various caryophyllene concentrations was prepared.

Background: The Greater Mekong Subregion (GMS) aims to eliminate all human malaria by 2030 and is making substantial progress toward this goal, with malaria increasingly confined to forest foci. These transmission foci are predominantly inhabited by ethnic minorities, local populations, and rural mobile and migrant populations working in mining and agriculture. The recommendations of the World Health Organization (WHO) on malaria elimination states that small population groups which constitute a large proportion of the malaria transmission reservoir should benefit from targeted strategies to reduce transmission overall.

While methyl-tertiary butyl ether (MTBE) remains the sole clinical topical agent for gallstone dissolution, its utility is limited due to side effects, largely stemming from its relatively low boiling point (55°C). In this study, we introduced 2-methoxy-6-methylpyridine (MMP), a novel gallstone-dissolving compound featuring an aromatic moiety and a substantially higher boiling point (156°C), designed to mitigate these side effects. We conducted a comprehensive evaluation of the efficacy and potential toxicities of MMP compared to MTBE using both in vitro and in vivo models.
