Amides from indole-3-glyoxylic acid and 4-benzoyl-2-methylpiperazine, which are related to entry inhibitors developed by Bristol-Myers Squibb (BMS), have been synthesized with aliphatic chains located at the C7 position of the indole ring. These spacers contain an azido group suitable for the well-known Cu(I)-catalyzed (3+2)-cycloaddition or an activated triple bond for the nucleophilic addition of thiols under physiological conditions. Reaction with polyols (β-cyclodextrin and hyperbranched polyglycerol) decorated with complementary click partners has afforded polyol-BMS-like conjugates that are not cytotoxic (TZM.bl cells) and retain the activity against R5-HIV-1 isolates. Thus, potential vaginal microbicides based on entry inhibitors, which can be called of 4 generation, are reported here for the first time.

In this study, a novel phenomenon is identified where precise control of topology and generation of polyglycerol induce the retention of Na ions in biological buffer systems, effectively inhibiting ice crystal growth during cryopreservation. Unlike linear and hyperbranched counterparts, densely-packed hydroxyl and ether groups in 4th-generation dendritic polyglycerol interact with the ions, activating the formation of hydrogen bonding at the ice interface. By inhibiting both intra- and extracellular ice growth and recrystallization, this biocompatible dendritic polyglycerol proves highly effective as a cryoprotectant; hence, achieving the cell recovery rates of ≈134-147%, relative to those of 10% dimethyl sulfoxide, which is a conventional cryoprotectant for human tongue squamous carcinoma (HSC-3) cell line and human umbilical vein endothelial (HUVEC) cells.

In this study, segmented hyperbranched copolymers with degradable and chain extendable cross-linker branch points were synthesized via green light-activated photoiniferter copolymerization of poly(ethylene glycol) methyl ether methacrylate (PEGMA) and a trithiocarbonate-derived dimethacrylate. A series of segmented hyperbranched copolymers with different degrees of branching were synthesized by changing the feed ratio of PEGMA to cross-linker to chain transfer agent. The segmented hyperbranched copolymers could be degraded into linear polymer chains by removing the trithocarbonate groups, which provides fundamental insights into the growth of primary chains during photoiniferter copolymerization.
