Aquaporins in platelet function.

Platelets

Structurally, aquaporins (AQPs) are small channel proteins with monomers of ~ 30 kDa that are assembled as tetramers to form pores on cell membranes. Aquaporins mediate the conduction of water but at times also small solutes including glycerol across cell membranes and along osmotic gradients. Thirteen isoforms of AQPs have been reported in mammalian cells, and several of these are likely expressed in platelets. Osmotic swelling mediated by AQP1 sustains the calcium entry required for platelet phosphatidylserine exposure and microvesiculation, through calcium permeable stretch-activated or mechanosensitive cation channels. Notably, deletion of AQP1 diminishes platelet procoagulant membrane dynamics and arterial thrombosis , independent of platelet granule secretion and without affecting hemostasis. Water entry into platelets promotes procoagulant activity, and AQPs may also be critical for the initiation and progression of venous thrombosis. Platelet AQPs may therefore represent valuable targets for future development of a new class of antithrombotics, namely, anti-procoagulant antithrombotics, that are mechanistically distinct from current antithrombotics. However, the structure of AQPs does not make for easy targeting of these channels, hence they remain elusive drug targets. Nevertheless, thrombosis data in animal models provide compelling reasons to continue the pursuit of AQP-targeted antithrombotics. In this review, we discuss the role of aquaporins in platelet secretion, aggregation and procoagulation, the challenge of drugging AQPs, and the prospects of targeting AQPs for arterial and venous antithrombosis.

Neurovascular unit, neuroinflammation and neurodegeneration markers in brain disorders.

Front Cell Neurosci

October 2024

Dr. Kiran C. Patel College of Osteopathic Medicine, Institute for Neuro-Immune Medicine, Nova Southeastern University, Ft. Lauderdale, FL, United States.

Article Synopsis
  • - The neurovascular unit (NVU) inflammation from glial cell activation and neuronal damage significantly contributes to neurodegenerative diseases, though the exact disease mechanisms are not fully understood.
  • - Biomarkers like neurofilament light (NfL) and glial fibrillary acidic protein (GFAP) can indicate the severity, progression, and treatment response in various brain disorders by assessing brain cell health and blood-brain barrier integrity.
  • - Chronic inflammation is prevalent in age-related neurodegenerative diseases such as Alzheimer's and Parkinson's, and certain biomarkers may change years before disease onset, highlighting their potential for early detection and research into neurovascular pathologies.

Role of eicosanoids in insect immunity: new insights and recent advances.

Insect Sci

August 2024

Post Graduate Department of Zoology, Banwarilal Bhalotia College, Asansol, Paschim Bardhaman, West Bengal, India.

Viruses, bacteria, fungus, protozoans, and different metazoan parasites and parasitoids present a constant threat to insects. Insect immunity has two components: humoral and cell mediated. Humoral immunity can be achieved by various antimicrobial proteins, namely, cecropins, sarcotoxin, defensin, attacin, etc.

Neuromyelitis optica spectrum disorder (NMOSD) is a rare autoimmune inflammatory demyelinating disease of the central nervous system affecting the optic nerves and spinal cord. Immune thrombocytopenia (ITP), on the other hand, is an autoimmune disorder characterized by a platelet count of <100 in the absence of any known condition that could be associated with thrombocytopenia. This case report focuses on a 56-year-old female presenting with the unique coexistence of NMOSD and ITP.

May 2024

Division of Pediatric Neurology, Department of Pediatrics I, Medical University of Innsbruck, Innsbruck, Austria. Electronic address:

Article Synopsis
  • The study focuses on comparing blood parameters in children with myelin oligodendrocyte glycoprotein antibody-associated disorders (MOGAD), AQP4-IgG-positive neuromyelitis optica spectrum disorders (NMOSD), multiple sclerosis (MS), and healthy controls to better understand disease activity.
  • Researchers analyzed complete blood counts and various ratios (like NLR and PLR) during different disease phases: clinical attack, acute treatment, and remission across a cohort of 174 children and adolescents.
  • Key findings revealed elevated leukocyte counts in MOGAD during clinical attacks, while other blood parameters like NLR and PLR showed different trends, suggesting these measures could aid in assessing disease activity and distinguishing between disease

Introduction: Neonatal stress disrupts brain development and increases the risk of neurological disorders later in life. However, the impact of neonatal stress on the development of the glymphatic system and susceptibility to Parkinson's disease (PD) remains largely unknown.

Methods: Neonatal maternal deprivation (NMD) was performed on mice for 14 consecutive days to model chronic neonatal stress.
