Programmable and modular systems capable of orthogonal genomic and transcriptomic perturbations are crucial for biological research and treating human genetic diseases. Here, we present the minimal versatile genetic perturbation technology (mvGPT), a flexible toolkit designed for simultaneous and orthogonal gene editing, activation, and repression in human cells. The mvGPT combines an engineered compact prime editor (PE), a fusion activator MS2-p65-HSF1 (MPH), and a drive-and-process multiplex array that produces RNAs tailored to different types of genetic perturbation.

One of the key processes that forms the basis of fertilisation is the tight interaction between sperm and egg. Both sperm and egg proteomes are known to evolve and diverge rapidly even between closely related species. Understanding the sperm proteome therefore provides key insights into the proteins that underpin the mechanisms involved during fertilisation and the fusion between sperm and egg, and how they can differ across individuals of the same species.

Capicua transcriptional repressor (CIC)-rearranged sarcoma (CRS) is a rare and recently described tumor that most commonly affects patients between 15 and 30 years of age. It is an undifferentiated round cell malignancy, with a disease defining CIC fusion, with double homeobox 4 (DUX4) being the most common partner. Here, we report a 77-year-old woman who presented with a cutaneous thigh mass with a clinical morphology suggesting Merkel cell carcinoma.

Background: In poultry, despite intense vaccination programs for prevention of Newcastle disease (ND), the ND infection still affects, causing high mortality in most vaccinated flocks.

Aim: This study aimed to determine whether the genetic material of the ND virus has changed and has become incompatible with the vaccines used in Iraq.

Methods: Real-time PCR was used to analyze genetic variation in the fusion (F) and haemaggluatination neuraminidase (HN) genes, as well as mRNA expression changes in inflammatory biomarkers, including C-reactive protein (CRP), interleukin 6, interleukin-1 beta (IL-6, IL-1β), and gamma interferon (IFN-γ).
