Novel flavonol-3--methylethers from Griseb. (Rutaceae).

Griseb. is a tree endemic to Cuba, occasionally used in herbal medicine. Previously, the antitrypanosomal activity of a -hexane-2-butanone extract of leaves and of its constituent skimmianine were published. In the current study a more thorough examination of the respective extract is performed, which led to the isolation and identification of three flavonoids, more specifically, the flavonol-3--methylethers kaempferol-3--methylether () and novel compounds kaempferol-3--methylether-5---D-glucoside () and kaempferol-8-hydroxy-3,7--dimethylether-5---D-glucoside (). All compounds were screened for their antimicrobial and antiprotozoal activity and cytotoxicity towards MRC-5 SV2 cells. Compound showed a moderate to weak activity against (IC 30.8 μM), (IC 15.4 μM) and (IC 53.8 μM), but also showed cytotoxicity (CC 19.0 μM). A novel silver-catalyzed cascade radical isonitrile insertion and defluorinative cyclization have been developed to synthesize CFH- and phosphinoyl-containing quinolines from -isocyanyl α-trifluoromethylstyrenes. The reaction proceeded under redox-neutral conditions and allowed the construction of a highly attractive quinoline ring system, with the simultaneous formation of the CFH group and introduction of various phosphinoyl groups in a single transformation, showing operational simplicity, a wide substrate scope, good tolerance for functional groups, and remarkable atom-/stepeconomy. Mechanistic studies indicated that the reaction is likely to involve the participation of P-centered radicals and key carbanion intermediates.
