Diphenyl phosphate (DPHP) has been increasingly detected in environmental samples, posing a potential hazard to humans and other organisms and arousing concern regarding its adverse effects. Biological degradation of DPHP is considered a promising and environmentally friendly method for its removal. In this study, the gene was mined from the W3 genome and identified as a glycerophosphodiester phosphodiesterase by bioinformatics analysis. The enzyme was expressed and its biochemical properties were studied. When using bis(4-nitrophenyl) phosphate as substrate, enzyme activity was optimal at 55 °C and a pH of 8.5. The enzyme remained stable in the pH range of 8.0 - 10.0. The rBaGDPD enzyme degraded DPHP and the reaction product was identified as phenyl phosphate by LC-MS. This is the first report of a glycerophosphodiester phosphodiesterase exhibiting hydrolytic activity against DPHP. This study demonstrated that rBaGDPD could have the potential for bioremediation and industrial applications.

Glycosylphosphatidylinositol (GPI)-anchored proteins (APs) regulate numerous biological processes through interaction with signaling effectors at the cell surface. As a unique feature, GPI-APs can be released from their anchors by multi-pass GPI-specific phospholipases (types A2, C, and D) to impact signaling networks, phenotype, and cell fate; however, many questions remain outstanding. Here, we discuss and expand our current understanding of the distinct GPI-specific phospholipases, their substrates, effector pathways, and emerging physiological roles, with a focus on the six-transmembrane ecto-phospholipases GDE2 (GDPD5) and GDE3 (GDPD2).

Aim: Breast cancer (BC) is the most frequently diagnosed malignancy worldwide, necessitating continued research into its molecular mechanisms. Circular RNAs (circRNAs) are increasingly recognized for their role in various cancers, including BC. This study explores the role of circRNA kinesin family member 4A (circKIF4A) in BC progression and its underlying molecular mechanisms.

Liquid chromatography tandem mass spectrometry (LC-MS/MS) is an essential tool for drug discovery that enables simple and rapid identification and quantification of chemical substances. A combination of a mobile phase and column makes it possible to analyze a wide range of target substances from low-molecular weight high-polar substances, such as amino acids and peptides, to low-polar substances such as lipids, and even macromolecular substances such as proteins. In this paper, we describe the results of applying LC-MS/MS to the analysis of phospholipids and related substances in biological samples and the analysis of photoproducts of pharmaceuticals.

Article Synopsis
  • Glycerophosphocholine (GPC) is a metabolite related to choline in cells and has been considered a potential supplement for brain health, but high choline intake may lead to TMAO production, linked to atherosclerosis.
  • This study investigates how GPC is absorbed and metabolized by the intestine, showing that it converts to choline and is transported into cells.
  • Researchers highlighted the enzyme Gpcpd1, which regulates this conversion and TMAO levels, with experiments indicating that its absence alters GPC metabolism and lowers TMAO in the bloodstream.

Article Synopsis
  • * Researchers collected 846 ticks from various environments and found a 6.2% overall infection rate of TBRFGB, identifying specific species such as Borrelia miyamotoi and Borrelia anserina in certain tick species.
  • * Additionally, a serological analysis of 42 patients with unexplained fevers showed that 10% had antibodies indicating possible recent infections with B. miyamotoi, highlighting the potential risk of these pathogens in the region.
