Background: In Alzheimer's disease (AD), changes in intestinal microbiota and systemic inflammation are concomitant with neuroinflammation and cognitive decline. This has led to the theory of microbial communities or infections as being causative in the development of neuroinflammation and immunosenescence seen in AD. Our research has demonstrated a decreased taxonomic diversity and an increased abundance of pathobionts in the gut of AD patients (Haran, mBio 2019), which is sufficient to promote amyloid and tau deposition in a mouse model (Chen, Gut 2023).

Background: Mosaic loss of the Y chromosome (LOY) is a somatic, age-related event that has been previously associated with a variety of diseases of aging. A prior study of European cohorts demonstrated an association between LOY and Alzheimer's Disease and more recent molecular studies have shown that LOY can also occur within microglia, suggesting a potential functional role in AD pathogenesis.

Method: In this study, we further validate the association between LOY in blood and AD via prospective analyses of 1,447 males.

Background: Homeostatic sleep regulation is theorized to be governed by the 'Two Process Model' where circadian rhythm (process C) and homeostatic sleep pressure (process S) interact to determine sleep versus wakefulness. Sleep pressure accumulated during prolonged wakefulness increases the duration and intensity of subsequent 'recovery' sleep. Multiple sleep abnormalities are associated with Alzheimer's Disease (AD) Dementia and Lewy Body Dementia (LBD).

Previous research indicates that both adults and newborns show enhanced electrophysiological and behavioral responses to schematic face-like configurations (FCs-three dots composing a downward-pointing triangle), as compared to the inverted configurations (ICs). Even fetuses, when exposed to light stimuli projected through the uterine wall, preferentially orient their heads toward FCs rather than ICs. However, when this effect emerges along the third trimester of pregnancy and in relation to the maturation of which brain structures is still unknown.
