Activation of autophagy is one of the responses elicited by high intraocular pressure (IOP) and mechanical stretch in trabecular meshwork (TM) cells. However, the mechanosensor and the molecular mechanisms by which autophagy is induced by mechanical stretch in these or other cell types is largely unknown. Here, we have investigated the mechanosensor and downstream signaling pathway that regulate cyclic mechanical stretch (CMS)-induced autophagy in TM cells. We report that primary cilia act as a mechanosensor for CMS-induced autophagy and identified a cross-regulatory talk between AKT1 and noncanonical SMAD2/3 signaling as critical components of primary cilia-mediated activation of autophagy by mechanical stretch. Furthermore, we demonstrated the physiological significance of our findings in ex vivo perfused eyes. Removal of primary cilia disrupted the homeostatic IOP compensatory response and prevented the increase in LC3-II protein levels in response to elevated pressure challenge, strongly supporting a role of primary cilia-mediated autophagy in regulating IOP homeostasis.

Molecular Clip Strategy of Modified Sulfur Cathodes for High-Performance Potassium Sulfur Batteries.

Adv Sci (Weinh)

January 2025

Joint Key Laboratory of the Ministry of Education, Institute of Applied Physics and Materials Engineering, University of Macau, Avenida da Universidade, Taipa, Macau SAR, 999078, China.

Potassium-sulfur (K-S) batteries are severely limited by the sluggish reaction kinetics of the cyclooctasulfur (cyclo-S) electrode with low conductivity, which urgently requires a novel cathode to facilitate activity to improve sulfur utilization. In this study, using the wet chemistry method, the molecular clip of Li is created to replace cyclo-S molecular with the highly active chain-like S molecular. The molecular clip strategy effectively lowers the reaction barrier in potassium-sulfur systems, and the stretching of S─S bonds weakens the binding between sulfur atoms, facilitating the transformation of potassium polysulfides (KPSs).

This study describes a complex human in vitro model for evaluating anti-inflammatory drug response in the alveoli that may contribute to the reduction of animal testing in the pre-clinical stage of drug development. The model is based on the human alveolar epithelial cell line Arlo co-cultured with macrophages differentiated from the THP-1 cell line, creating a physiological biological microenvironment. To mimic the three-dimensional architecture and dynamic expansion and relaxation of the air-blood-barrier, they are grown on a stretchable microphysiological lung-on-chip.

Incomplete tendon healing and postponed muscle weakness after Achilles tendon rupture and surgical repair lead to poor performance in patient activities. Although the effectiveness of postoperative early functional rehabilitation has been proven, the priority and each effect of specific methods in early rehabilitation remain unclear. We hypothesized early muscle contraction exercises without joint motion would promote tendon healing and prevent calf muscle atrophy; in contrast, early static stretching after surgical repair would not contribute to tendon healing and induce calf muscle atrophy.

Bioinspired Smart Triboelectric Soft Pneumatic Actuator-Enabled Hand Rehabilitation Robot.

Adv Mater

January 2025

Division of Intelligent and Biomechanical Systems, State Key Laboratory of Tribology in Advanced Equipment, Department of Mechanical Engineering, Tsinghua University, Haidian, Beijing, 100084, China.

Quantitative assessment for post-stroke spasticity remains a significant challenge due to the encountered variable resistance during passive stretching, which can lead to the widely used modified Ashworth scale (MAS) for spasticity assessment depending heavily on rehabilitation physicians. To address these challenges, a high-force-output triboelectric soft pneumatic actuator (TENG-SPA) inspired by a lobster tail is developed. The bioinspired TENG-SPA can generate approximately 20 N at 0.

Damage mechanisms are a key factor in materials science and are essential for understanding and predicting the behavior of materials under complex loading conditions. In this paper, the influence of different directions, different rates and different model parameters on the mechanical behavior of AZ31 magnesium alloy during the tensile process is investigated based on the secondary development of the VUMAT user subroutine based on the GTN damage model and verified by the tensile experiments at different loading rates and in different directions. The results show that AZ31 magnesium alloy exhibits significant differences in mechanical properties in radial and axial stretching, where the yield strength is lower in the radial direction than in the axial direction, and the elongation is the opposite.
