AI Article Synopsis

  • - Platinum(IV) complexes combined with orotic acid show effective targeting of liver cancer cells, particularly in Hep G2 cells, indicating higher activity and uptake.
  • - The increased activity is linked to the higher expression of Organic Anion Transporter 2 (OAT2) in Hep G2 cells, suggesting that OAT2 plays a key role in the uptake of these complexes, especially when the OAT2 inhibitor is present.
  • - Unlike traditional chemotherapy drugs like cisplatin and oxaliplatin, these platinum(IV) complexes are not hindered by the copper transporter ATP7B, making them more promising for treating liver cancer.

Article Abstract

Platinum(IV) complexes of orotic acid selectively target liver cancer cells displaying enhanced activity and higher uptake in Hep G2. The comparatively higher expression of Organic Anion Transporter 2 (OAT2) in Hep G2 and decrease in toxicity in the presence of OAT2 inhibitor suggest its involvement in the uptake of the complexes. They are resistant to sequestration by the copper transporter ATP7B, unlike cisplatin and oxaliplatin.

Green microalga conserves substrate uptake pattern but changes their metabolic uses across trophic transition.

Front Microbiol

November 2024

Environmental Genomics and Systems Biology, Lawrence Berkeley National Laboratory, Berkeley, CA, United States.

The terrestrial green alga is an emerging model species with potential applications including production of triacylglycerol or astaxanthin. How interacts with the diverse substrates during trophic transitions is unknown. To characterize its substrate utilization and secretion dynamics, we cultivated the alga in a soil-based defined medium in transition between conditions with and without glucose supplementation.

Gut Microbiota and Related Metabolites in Children With Egg White Sensitization.

Pediatr Infect Dis J

December 2024

Department of Biomedical Sciences, College of Medicine, Chang Gung University, Taoyuan, Taiwan.

Background: We hypothesized that food sensitization in children could be linked to specific gut microbiota. The objective of this study is to assess a group of children with egg white sensitization (ES) from the microbiological and biochemical-metabolic standpoint, applying the microbiota and metabolomics approach to studying the intestinal contents of the feces.

Methods: Twenty-eight toddlers with ES (mean age 13.

The identification of individuals with the greatest risk of progression to active tuberculosis (TB) disease from the huge reservoir of () infected individuals continues to remain an arduous ascent in the global effort to control TB. In a two-year prospective study, we analysed metabolic profiles in the unstimulated and TB antigen stimulated QuantiFERON supernatants of 14 healthy household contacts (HHCs) who progressed to TB disease (Progressors) and 14 HHCs who remained healthy (Non-Progressors). We identified 21 significantly dysregulated metabolites in the TB antigen-stimulated QuantiFERON supernatants of Progressors, of which the combination of Malic acid and N-Arachidonoylglycine had maximum AUC of 0.

School of Biotechnology and Key Laboratory of Carbohydrate Chemistry and Biotechnology of Ministry of Education, Jiangnan University, Wuxi 214122, China.

Article Synopsis
  • Cytidine 5'-monophosphate (5'-CMP) is an important compound for food and pharmaceutical applications, and its production has been improved in engineered strains of bacteria.
  • In the study, the engineered strain CM006 showed a 134.7-fold increase in 5'-CMP production by blocking its degradation pathway, and further modifications led to higher yields of the compound.
  • The highest reported titer of 5'-CMP reached 417.9 mg/L in strain CM012, demonstrating effective methods for sustainable industrial production of this valuable nucleotide.

Hyperammonemia is a serious metabolic condition marked by elevated ammonia levels in the blood, leading to neurological damage and systemic complications if untreated. While often associated with liver dysfunction, inborn metabolic errors such as fatty acid oxidation defects, pyruvate metabolism disorders, urea cycle disorders (UCDs), urea splitting bacterial infections, hemato-oncological disorders, and portosystemic shunts are less commonly recognized but significant causes, particularly outside neonatal populations. These metabolic errors, due to partial enzyme deficiencies, may present later in life with atypical symptoms.
