Correlative light and electron microscopy (CLEM) entails a group of multimodal imaging techniques that are combined to pinpoint to the location of fluorescently labeled molecules in the context of their ultrastructural cellular environment. Here we describe a detailed workflow for STORM-CLEM, in which STochastic Optical Reconstruction Microscopy (STORM), an optical super-resolution technique, is correlated with transmission electron microscopy (TEM). This protocol has the advantage that both imaging modalities have resolution at the nanoscale, bringing higher synergies on the information obtained. The sample is prepared according to the Tokuyasu method followed by click-chemistry labeling and STORM imaging. Then, after heavy metal staining, electron microscopy imaging is performed followed by correlation of the two images. The case study presented here is on intracellular pathogens, but the protocol is versatile and could potentially be applied to many types of samples.

Due to a lack of spatially resolved characterization studies on statistical and individual particle microstructure at multiple scales, a knowledge gap exists in understanding the mechanistic link between rapid performance failure and atomic-scale structure degradation in single-crystalline Ni-rich battery cathodes. In a recent publication in , Huang developed a multi-crystal rocking curve technique (combining X-ray and electron microscopy to capture both statistical and individual lattice distortions), which enables multiscale observations and further proves that the accumulation of the unrecoverable lattice rotation in cathodes upon repeated cycling exacerbates mechanical failure and electrochemical decay. The elucidation of failure mechanisms in single-crystalline cathodes offers valuable insights into the development of long-lasting and high-energy-density cathodes in next-generation batteries, encompassing strategies to mitigate lattice rotation and enhance lattice structure tolerance against lattice distortion within individual particles.

Tissue regeneration after a wound occurs through three main overlapping and interrelated stages namely inflammatory, proliferative, and remodelling phases, respectively. The inflammatory phase is key for successful tissue reconstruction and triggers the proliferative phase. The macrophages in the non-healing wounds remain in the inflammatory loop, but their phenotypes can be changed interactions with nanofibre-based scaffolds mimicking the organisation of the native structural support of healthy tissues.
