Classical circulating LyC6 murine monocytes differentiate progressively from inflammatory tissue monocytes to mature macrophages (Mϕ) after entry into gut mucosa. This protocol provides a two-step in vitro culture method that replicates the human monocyte maturation cascade. First, purified circulating CD14 CD16 monocytes exposed to granulocyte-macrophage colony-stimulating factor (GM-CSF), interferon gamma (IFNγ), and interleukin 23 (IL-23) differentiate into tissue-like inflammatory monocytes. Next, addition of transforming growth factor beta (TGFβ) plus interleukin 10 (IL-10) promotes their maturation into tissue-like Mϕ. Methods to sort these cells after culture are also provided. The fine-tuning of this system might open therapeutic avenues for chronic inflammatory disorders. © 2021 Wiley Periodicals LLC Basic Protocol 1: Isolation of human monocytes from peripheral blood Basic Protocol 2: First step culture for generation of inflammatory monocyte-like cells Basic Protocol 3: Second step culture for differentiation of inflammatory monocyte-like cells into macrophages Alternate Protocol: Sorting and culturing of inflammatory monocyte-like cells.

Vulvovaginal candidiasis (VVC) represents the second cause of vaginal infections in childbearing-age women. It mainly affects the vulva and vagina; however, other organs can be compromised, with consequences that are not well known yet. To evaluate the ability of Candida albicans, inoculated into the vaginal lumen of mice, to migrate to the uterus and ovaries.

Mesothelioma is a lethal cancer of the serosal lining of the body cavities. Risk factors include environmental and genetic factors. Asbestos exposure is considered the principal environmental risk factor, but other carcinogenic mineral fibers, such as erionite, also have a causal role.

In vivo calcium imaging in freely moving rats using miniscopes provides valuable information about the neural mechanisms of behavior in real time. A gradient index (GRIN) lens can be implanted in deep brain structures to relay activity from single neurons. While such procedures have been successful in mice, few reports provide detailed procedures for successful surgery and long-term imaging in rats, which are better suited for studying complex human behaviors.

Background: Although physical activity (PA) has been linked to cognitive health, the nuanced relationships between different dimensions of PA and cognitive impairment remain inconclusive. This study investigated associations between late-life PA levels, midlife-to-late-life activity patterns, and cognitive impairment in Chinese older adults, considering potential moderation by apolipoprotein E (APOE) ε4 genotype.
Methods: We analyzed baseline data from 6,899 participants (median age 68 years, 55.

The circadian clock genes are known important for kidney development, maturation and physiological functions. However, whether and how they play a role in renal regeneration remain elusive. Here, by using the single cell RNA-sequencing (scRNA-seq) technology, we investigated the dynamic gene expression profiles and cell states after acute kidney injury (AKI) by gentamicin treatment in zebrafish.
