A pH-sensitive nanoagent self-assembled from a highly negatively-charged phthalocyanine with excellent biosafety for photothermal therapy.

Photothermal therapy (PTT) is a promising strategy for cancer treatment. However, the development of highly efficient photothermal agents with excellent biosafety, particularly with low liver retention, is very meaningful for clinical applications, but it is also challenging. We herein prepared a pH-sensitive nanoagent (NanoPc3) by the self-assembly of a zinc(ii) phthalocyanine substituted with hexadeca-sulphonates linked by hydrazone bonds for photoacoustic imaging and PTT. Due to the highly negative surface potential (-30.80 mV in water), NanoPc3 could effectively escape the phagocytosis of the reticuloendothelial system and be rapidly cleared from normal tissues, leading to little accumulation in the liver and excellent biosafety. The highly negatively-charged NanoPc3 changed into nearly neutral nanoparticles (NanoPc3H) under slightly acidic conditions, resulting in enhanced cellular uptake and retention time in tumor tissues. Moreover, the tumor of H22 tumor-bearing mice treated with NanoPc3 almost disappeared, suggesting an outstanding photothermal antitumor effect. NanoPc3 also hardly showed skin phototoxicity under irradiation. Its excellent antitumor effect and biosafety make NanoPc3 highly promising in clinical applications. This work will provide a new strategy for the design of tumor-targeted photothermal nanoagents with high biosafety.

Immunocompromised populations, including cancer patients, elderly individuals, and those with chronic diseases, are the primary targets of superbugs. Traditional vaccines are less effective due to insufficient or impaired immune cells. Inspired by the "vanguard" effect of neutrophils (NE) during natural infection, this project leverages the ability of NE to initiate the NETosis program to recruit monocytes and DC cells, designing vaccines that can rapidly recruit immune cells and enhance the immune response.

The development of safe, environmentally friendly, edible antimicrobial packaging films represents a promising alternative to conventional plastic packaging for reducing spoilage and extending the shelf life of fresh food. Here, we propose a novel strategy to construct edible β-CD-MOF/carvacrol@zein (BCCZ) composite films by intertwining β-CD-MOF loaded with the antimicrobial essential oil carvacrol, and zein. The resulting BCCZ films exhibit high humidity-triggered, long-lasting bactericidal efficacy, effective fruit preservation, and excellent biosafety.

In this paper, a pH-sensitive chitosan-grafted phenylboronic acid (CS-BA)/polyvinyl alcohol (PVA) hydrogel was constructed based on dynamic borate bonding for loading chemotherapeutic drug cisplatin (CDDP) and divalent Cu (CS-BA/PVA-Cu-CDDP). The hydrogel can respond and degrade specifically in the simulative acidic tumor microenvironment (TME), and the released Cu can deplete glutathione (GSH) in tumor cells and generate Cu. It is worth noting that, Cu can further catalyze the Fenton-like reaction to generate cancer cell-toxic hydroxyl radicals (OH•).
