Application of LC-MS/MS-based molecular networking indicated the ethanol extract of octocoral is a potential source for the discovery of new xenicane derivatives. A natural product investigation of this soft coral resulted in the isolation of four new xenicane diterpenoids, asterolaurins O‒R (‒), together with six known compounds, xeniolide-A (), isoxeniolide-A (), xeniolide-B (), 7,8-epoxyxeniolide-B (), 7,8-oxido-isoxeniolide-A (), and 9-hydroxyxeniolide-F (). The structures of isolated compounds were characterized by employing spectroscopic analyses, including 2D-NMR (COSY, HMQC, HMBC, and NOESY) and high-resolution electrospray ionization mass spectrometry (HRESIMS). Asterolaurin O is the first case of brominated tricarbocyclic type floridicin in the family Xeniidae. Concerning bioactivity, the cytotoxic activity of those isolates was evaluated. As a result, compounds and demonstrated a selective cytotoxic effect against the MCF-7 cell line at IC of 14.7 and 25.1 μM, respectively.

Anti-inflammatory diterpenoids from the brown alga Dictyota coriacea.

Biosci Biotechnol Biochem

November 2024

Department of Chemistry, Graduate School of Science, Osaka Metropolitan University, Osaka, Japan.

A new xenicane diterpene named 4α-acetoxyisodictyohemiacetal (1) was isolated from the brown alga Dictyota coriacea, along with 11 known diterpenoids. The structure of 1 was established by spectroscopic analyses, and its absolute configuration was determined by comparing the experimental and theoretical electronic circular dichroism (ECD) spectra. Two dictyodiacetal diastereomers (5 and 6) were isolated and the full NMR assignments were performed.

Waixenicin A, a xenicane diterpene from the octocoral , is a selective, potent inhibitor of the TRPM7 ion channel. To study the structure-activity relationship (SAR) of waixenicin A, we isolated and assayed related diterpenes from . In addition to known waixenicins A () and B (), we purified six xenicane diterpenes, 7,8-epoxywaixenicins A () and B (), 12-deacetylwaixenicin A (), waixenicin E (), waixenicin F (), and 20-acetoxyxeniafaraunol B ().

A phytochemical investigation of an East China Sea collection of the brown alga has led to the isolation of four novel nitrogen-containing crenulide diterpenoids, named coriaceumins A-D (-), two rare nitrogenous xenicane diterpenoids, dictyolactams C () and D (), and one known crenulide diterpenoid, hydroxycrenulide (). The structures of the new compounds were elucidated by detailed spectroscopic data analyses, including HRESIMS and 1D/2D NMR. The absolute configurations were determined by a comparison of the experimental ECD spectra with the spectra computed by DFT-based quantum chemical calculations.

February 2023

School of Pharmaceutical Sciences, Wenzhou Medical University, Wenzhou, Zhejiang 325035, People's Republic of China.

Five new xenicane diterpenes, including three rare nitrogen-containing derivatives, dictyolactams A () and B () and 9-demethoxy-9-ethoxyjoalin (), a rare diterpene with a cyclobutanone moiety, named 4-hydroxyisoacetylcoriacenone (), and 19--acetyldictyodiol (), were isolated from an East China Sea collection of the brown alga , along with 15 known analogues (-). The structures of the new diterpenes were elucidated by spectroscopic analyses and theoretical ECD calculations. All compounds had cytoprotective effects against oxidative stress in neuron-like PC12 cells.
