We describe two complementary approaches based on a convergent [4+2] logic toward the synthesis of amorfrutins, cannabinoids, and related plant metabolites. An anionic cascade cyclization employing β-methoxycrotonates and β-chloro-α,β-unsaturated esters yielded amorfrutins in four linear steps and demonstrated utility of β-alkoxycrotonate-derived nucleophiles as functional equivalents of β-ketoester-derived dianions. Analogously, tandem Diels-Alder/retro-Diels-Alder cycloaddition of dimedone-derived bis(trimethylsiloxy)-dienes and α,β-alkynyl ester dienophiles provided facile access to resorcinol precursors of amorfrutins and cannabinoids, avoiding late-stage installation of prenyl or geranyl moieties as in previous approaches.

A dual-mode detection platform utilizing colorimetric and Raman was developed based on the exponential amplification reaction (EXPAR) strategy and a "core-satellite" structure constructed by bimetallic nanozymes to detect chloramphenicol (CAP). Initially, DNA-gated metal-organic frameworks (MOFs) incorporating cascaded amplification were used to be nanocarriers for the colorimetric and Raman reporter molecules (3,3',5,5'-tetramethylbiphenyl; TMB). Subsequently, assembled DNA served as gatekeepers to create a stimulus-responsive DNA-gated MOF (TMB@DNA/MOF).

Severe traumatic bleeding and chronic diabetic wounds require rapid hemostasis and multifunctional dressings, which remain particularly challenging, especially for non-compressible trauma and irregular wounds with dysregulated microenvironments. Chitosan (CS) can be easily cross-linked with genipin to form GpCS hydrogels. Chemodynamic therapy (CDT) is a promising and potent therapeutic strategy for the treatment of cancer. We developed a DNA origami-based enzymatic cascade nanoreactor (DOECN) containing spatially well-organized Au nanoparticles and ferric oxide (FeO) nanoclusters for targeted delivery and inhibition of tumor cell growth. The DOECN can synergistically promote the generation of hydrogen peroxide (HO), consumption of glutathione, and creation of an acidic environment, thereby amplifying the Fenton-type reaction and producing abundant reactive oxygen species, such as hydroxyl radicals (•OH), for augmenting the CDT outcome.
