Visible-light-driven functionalization of various organic systems has proved to be extremely successful and reached an impressive level of sophistication as well as efficiency in the last two decades. At the same time, organosilicon compounds are significant due to their promising applications in therapeutic agents, drug delivery, building blocks and so on. More interestingly, they are cheap, operationally simple, highly stable, less toxic and easy to handle. In this scenario, an environment-friendly synthetic approach for silylation, such as visible-light-induced silylation, is in high demand at present for molecule construction having the C-Si bond. This perspective summarizes the recent findings and developments in the emerging area of photocatalytic silylation with literature coverage mainly extending from 2014 to February 2021.

Total Syntheses of Bryostatins 1, 7, 9 and 9-N3.

Bryostatins are a family of marine natural products that have garnered significant interests, as evidenced by over 40 clinical trials. However, their extremely low natural abundance has severely limited further research. Despite significant efforts, which have led to the total synthesis of seven bryostatin members by eight independent research groups, these complex molecules present persistent challenges for stereocontrolled, large-scale, and especially divergent synthesis.

A visible-light-induced synthesis protocol for silylmonofluoroalkanes is described. The silylation of alkenyl fluorides using (trimethylsilyl)silanes as organosilicon reagents proceeds well under mild conditions via a sequential photoinduced single-electron transfer and protonation process. The protocol shows a broad substrate scope, transition-metal-free conditions, and high functional group tolerance.

Photoinduced halogen-atom transfer (XAT) has rapidly emerged as a programmable approach to generate carbon-centered radical intermediates, mainly relying on silyl and α-aminoalkyl radicals as halogen abstractors. More recently, ligated boryl radicals have also been proposed as effective halogen abstractors under visible-light irradiation. In this study, we describe the use of this approach to enable C(sp)-C(sp) bond formation radical addition of carbon-centered radicals generated XAT onto chloroalkynes.

Iron-Catalyzed Electrophotochemical α-Functionalization of a Silylcyclobutanol.

Org Lett

State Key Laboratory of Elemento-Organic Chemistry, Research Institute of Elemento-Organic Chemistry, College of Chemistry, Frontiers Science Center for New Organic Matter, Nankai University, Tianjin 300071, China.

Four-membered ring structure is important in organic chemistry, and selective cleavage and functionalization of these strained rings are of great interest. However, direct α-functionalization of cyclobutanols is rarely reported because of the high O-H bond dissociation energy and the occurrence of β-scission of C-C bonds in these alcohols. Recently, transition-metal catalysis has facilitated alkoxy radical generation.

Stereoselective Synthesis of Silyl Enol Ethers with Acylsilanes and α,β-Unsaturated Ketones.

Acylsilanes are emerging bench-stable reagents for the generation of electron-rich oxycarbenes that are difficult to access with unstable diazo compounds. Herein, we report a siloxycarbene-mediated stereoselective synthesis of silyl enol ethers through visible-light-induced intermolecular reactions between acylsilanes and α,β-unsaturated ketones. Both the solvent and low temperature are important for the success of the reaction.
