Ratiometric pH sensing by fluorescence resonance energy transfer-based hybrid semiconducting polymer dots in living cells.

Intracellular pH plays a significant role in all cell activities. Due to their precise imaging capabilities, fluorescent probes have attracted much attention for the investigation of pH-regulated processes. Detecting intracellular pH values with high throughput is critical for cell research and applications. In this work, hybrid semiconducting polymer dots (Pdots) were developed and characterized and were applied for cell imaging and exclusive ratiometric sensing of intracellular pH values. The reported Pdots were prepared by blending a synthesized block polymer (P) and a semiconducting polymer poly[2-methoxy-5-(2-ethylhexyloxy)-1,4-phenylenevinylene] (MEHPPV) to construct a fluorescence resonance energy transfer system for ratiometric sensing. Pdots showed many advantages, including high brightness, excellent photostability and biocompatibility, giving the pH probe high sensitivity and good stability. Our results proved the capability of P-MEHPPV Pdots for the detection of pH in living cells.

A new Rhodamine functionalised Schiff Base sensor 3',6'-bis(diethylamino)-2-((4-hydroxybenzylidene)amino)spiro[isoindoline-1,9'-xanthen]-3-one (SBRB1) was designed and synthesized. The recognition ability of sensor SBRB1 towards Hg was studied by using UV-Vis and fluorescence spectroscopy. The fluorescence results showed that the sensor SBRB1 has specific selectivity as well as sensitivity towards Hg among other competitive metal ions as the fluorescence intensity at 479 nm quenched only in the presence of Hg.

A ratiometric fluorescent nanoprobe (CDs-Rho), synthesized through the simple covalent amide linkage between carbon dots (CDs) and pH-sensitive rhodamine dye (Rho), was designed for the precise sensing and imaging of extremely alkaline environments. The sensing mechanism involves the opposite pH-dependent fluorescence changes in CDs and Rho, respectively, coupled with pH-regulated FRET efficiency from CDs to Rho. The extensive use of tetracyclines in livestock poses health risks due to their residues in animal-derived food; therefore, developing simple detection methods to replace complex traditional approaches is of paramount importance. Here, we developed a dual-ligand zinc-based metal-organic framework material, Zn-BTC-BDC-NH (denoted as ZTD), for the detection of tetracyclines. The intrinsic blue fluorescence of ZTD was quenched upon the introduction of tetracyclines due to electron transfer from -NH of ZTD to -CO- and -OH groups of tetracycline molecules; meanwhile, the new green fluorescence emission was generated through π-π stacking between aromatic rings and the formation of complexes between Zn and C-O/C═O groups.
