We evaluated the potential for a monoclonal antibody antagonist of the glucagon receptor (Ab-4) to maintain glucose homeostasis in type 1 diabetic rodents. We noted durable and sustained improvements in glycemia which persist long after treatment withdrawal. Ab-4 promoted β-cell survival and enhanced the recovery of insulin islet mass with concomitant increases in circulating insulin and C peptide. In PANIC-ATTAC mice, an inducible model of β-cell apoptosis which allows for robust assessment of β-cell regeneration following caspase-8-induced diabetes, Ab-4 drove a 6.7-fold increase in β-cell mass. Lineage tracing suggests that this restoration of functional insulin-producing cells was at least partially driven by α-cell-to-β-cell conversion. Following hyperglycemic onset in nonobese diabetic (NOD) mice, Ab-4 treatment promoted improvements in C-peptide levels and insulin islet mass was dramatically increased. Lastly, diabetic mice receiving human islet xenografts showed stable improvements in glycemic control and increased human insulin secretion.

Methicillin-resistant (MRSA) causes osteomyelitis (OM), which seriously threatens public health due to its antimicrobial resistance. To increase the sensitivity of antibiotics and eradicate intracellular bacteria, a Zn and vancomycin (Van) codelivered nanotherapeutic (named Man-Zn/Van NPs) was fabricated and characterized via mannose (Man) modification. Man-Zn/Van NPs exhibit significant inhibitory activity against extra- and intracellular MRSA and obviously decrease the minimum inhibitory concentration of Van.

Rapid reduction of body size in populations responding to global warming suggests the involvement of temperature-dependent physiological adjustments during growth, such as mitochondrial alterations, in the efficiency of producing metabolic energy, a process that is poorly explored, especially in endotherms. Here, we examined the mitochondrial metabolism and proteomic profile of red blood cells in relation to body size and cellular energetics in nestling shearwaters (Calonectris diomedea) developing at different natural temperatures. We found that nestlings of warmer nests had lighter bodies and smaller beaks at fledging.

Retroperitoneal lymphangioma is exceptionally rare. We present a case of a 41-year-old asymptomatic patient with a large abdominal cystic mass detected on contrast-enhanced computed tomography (CT) scan, initially suspected to be pseudomyxoma peritonei. Laparoscopic exploration revealed a 30 x 30 cm multilocular cystic tumour originating from the retroperitoneum.
