Surface-enhanced Raman scattering (SERS) spectra contain information on the chemical structure on nanoparticle surfaces through the position and alignment of molecules with the electromagnetic near field. Time-dependent density functional theory (TDDFT) can provide the Raman tensors needed for a detailed interpretation of SERS spectra. Here, the impact of molecular conformations on SERS spectra is considered. TDDFT calculations of the surfactant cetyltrimethylammonium bromide with five conformers produced more accurate unenhanced Raman spectra than a simple all-trans structure. The calculations and measurements also demonstrated a loss of structural information in the CH/CH scissor vibration band at 1450 cm in the SERS spectra. To study lipid bilayers, TDDFT calculations on conformers of methyl phosphorylcholine and -5-decene served as models for the symmetric choline stretch in the lipid headgroup and the C═C stretch in the acyl chains of 1,2-oleoyl--3-phosphocholine. Conformer considerations enabled a measurement of the distribution of double-bond orientations with an order parameter of = 0.53.

The heterogeneity of extracellular vesicles (EVs) surface information represents different functions, which is neglected in previous studies. In this study, a label-free SERS analysis approach is demonstrated to study fundamental EV biological and physical information heterogeneity by matching specific sizes of nano-enhanced particles. This strategy reveals informative, comprehensive, and high-quality SERS spectra of the overall exosome surface, and effectively circumvents the key information loss caused by the spatial resistance of NPs binding to the 293 exosomes' concave structure.

The complexity of serum constituents and the lack of knowledge on biomarkers bring obstacles for reliable cancer screening with the limited information obtained by surface-enhanced Raman scattering (SERS) on a single substrate. We report here an extremely high-accurate approach for cancer screening by biomarker-free serum-SERS technique which employs three principal SERS substrates that are of specially designed surface properties. With three substrates rich information on serum can be obtained from SERS spectra, of which quadratic discriminant analysis (QDA) yields excellent classifications of serum samples independently.

Here we show how surface-enhanced Raman spectroscopy (SERS) features can be fine-tuned in optically active substrates made of layered materials. To demonstrate this, we used DNA-assisted lithography (DALI) to create substrates with silver bowtie nanoparticle-aperture pairs and then coated the samples with rhodamine 6G (R6G) molecules. By varying the spacing between the aperture and particle layer, we were able to control the strength of the interlayer coupling between the plasmon resonances of the apertures and those of the underlying bowtie particles.
