Small angle neutron scattering (SANS) is a powerful tool for studying the structure of solubilized membrane proteins. It allows describing the general dimension of the membrane protein , evidence conformational changes, and may provide a low-resolution structure at the nm resolution range. This is because SANS can discriminate between the membrane protein and its amphiphilic partner by specific deuteration of the partners and of the buffer. This chapter was written to offer to a scientist aiming to describe a membrane protein structure the basic tools to consider a SANS experiment. It presents the general principle of contrast variation and a bibliographic survey of experimental strategies used for membrane proteins, some basic theoretical background, and a succinct description of the principles of analysis, of the instrumental and sample requirement, and of the practical steps, prior to the experiments, during the experiments and for data analysis.

Itch is a dominant symptom in dermatitis, and scratching promotes cutaneous inflammation, thereby worsening disease. However, the mechanisms through which scratching exacerbates inflammation and whether scratching provides benefit to the host are largely unknown. We found that scratching was required for skin inflammation in mouse models dependent on FcεRI-mediated mast cell activation.

Objective: To investigate the effects and mechanisms of miRNA 221 on myocardial ischemia/reperfusion injury (MIRI) in mice through the regulation of phospholamban (PLB) expression.
Methods: The MIRI mouse model was created and mice were divided into sham, MIRI, MIRI+ 221, and MIRI+ scr groups, with miRNA 221 overexpression induced in the myocardium of MIRI mice by targeted myocardial injection. Quantitative RT-PCR analysis was performed to observe the variation in miRNA 221, PLB, SERCA2, RYR2, NCX1, Cyt C and caspase 3 mRNA levels in myocardium, while Western blot assessed the levels of PLB, p-PLB (Ser16), p-PLB (Thr17), SERCA2, RYR2, NCX1, Cyt C and caspase 3 proteins.
