Open searching has proven to be an effective strategy for identifying both known and unknown modifications in shotgun proteomics experiments. Rather than being limited to a small set of user-specified modifications, open searches identify peptides with any mass shift that may correspond to a single modification or a combination of several modifications. Here we present PTM-Shepherd, a bioinformatics tool that automates characterization of post-translational modification profiles detected in open searches based on attributes, such as amino acid localization, fragmentation spectra similarity, retention time shifts, and relative modification rates. PTM-Shepherd can also perform multiexperiment comparisons for studying changes in modification profiles, e.g., in data generated in different laboratories or under different conditions. We demonstrate how PTM-Shepherd improves the analysis of data from formalin-fixed and paraffin-embedded samples, detects extreme underalkylation of cysteine in some data sets, discovers an artifactual modification introduced during peptide synthesis, and uncovers site-specific biases in sample preparation artifacts in a multicenter proteomics profiling study.

Bone marrow aspiration is an essential procedure for diagnosing hematological conditions and therapeutic applications, such as bone marrow aspirate concentrate (BMAC) therapy for non-union of bones. This report presents a case of a 35-year-old male with femoral non-union treated with BMAC, requiring the collection of a large bone marrow aspirate volume. A novel, low-cost modification to the standard bone marrow aspiration apparatus was developed using components from an intravenous (IV) drip set, including a rubber bulb connector and a luer lock connector.

Gallstone disease during pregnancy, or cholelithiasis, presents significant clinical challenges due to hormonal, anatomical, and metabolic changes. Progesterone therapy, commonly used in pregnancy for uterine bleeding, can exacerbate gallstone risk by reducing gallbladder motility and promoting cholesterol gallstone formation. This case report describes a 29-year-old pregnant woman with no prior gallbladder disease who developed multiple cholesterol gallstones during the third trimester while undergoing progesterone therapy for bleeding associated with a bicornuate uterus.

The rational design of non-fullerene acceptors (NFAs) with both high crystallinity and photoluminescence quantum yield (PLQY) is of crucial importance for achieving high-efficiency and low-energy-loss organic solar cells (OSCs). However, increasing the crystallinity of an NFA tends to decrease its PLQY, which results in a high non-radiative energy loss in OSCs. Here we demonstrate that the crystallinity and PLQY of NFAs can be fine-tuned by asymmetrically adapting the branching position of alkyl chains on the thiophene unit of the L8-BO acceptor.

Background: This qualitative evaluation was embedded in the Rehabilitation Exercise and psycholoGical support After COVID-19 InfectioN (REGAIN) study, a randomised controlled trial (RCT) for those with post-COVID-19 condition ('long COVID') after hospital admission for COVID-19, comparing weekly home-based, live online supervised group exercise and psychological support sessions with 'best practice usual care' (a single session of advice).
