Human Paraoxonase-2 (PON2): Protein Functions and Modulation.

Published: February 2021

PON1, PON2, and PON3 belong to a family of lactone hydrolyzing enzymes endowed with various substrate specificities. Among PONs, PON2 shows the highest hydrolytic activity toward many acyl-homoserine lactones (acyl-HL) involved in bacterial quorum-sensing signaling. Accordingly, defense against pathogens, such as (), was postulated to be the principal function of PON2. However, recent findings have highlighted the importance of PON2 in oxidative stress control, inhibition of apoptosis, and the progression of various types of malignancies. This review focuses on all of these aspects of PON2.

Deepening Cisplatin sensitivity on Oral Squamous cell Carcinoma cell lines after PON2 knockdown: A FTIRM investigation.

Cisplatin is a platinum-based chemotherapy drug with antimicrobial and antitumoral activity, largely used for a long time in the treatment of several cancers, including the Oral Squamous Cell Carcinoma (OSCC), which is one of the most frequent neoplasms of the oral cavity. Due to its aggressiveness and metastatic invasion, OSCC is characterized by poor outcome, often related also to chemoresistance mechanisms. The intracellular enzyme paraoxonase-2 (PON2) normally acts defending cells from the damages induced by Reactive Oxygen Species.

Myocardial infarction (MI) is a multifactorial polygenic disease that develops as a result of a complex interaction of numerous genetic factors and the external environment. Accordingly, the contribution of each of them separately is usually not large and may significantly depend on the state of other accompanying factors. The purpose of the study was to search for informative predictors of MI risk based on polygenic analysis of polymorphic variants of (1) the antioxidant defense enzyme genes PON1 (rs662), PON2 (rs7493), CAT (rs1001179), MSRA (rs10098474) and GSTP1 (rs1695); (2) the apoptosis genes CASP8 (rs3834129), TP53 (rs1042522) and BCL2 (rs12454712); and (3) the inflammation genes CRP (rs1205), CX3CR1 (rs3732378), IL6 (rs1800795) and CCL2 (rs1024611).

Immediate Postoperative Changes After Expansion Pharyngoplasty and Hypoglossal Nerve Stimulation.

Laryngoscope

November 2024

Department of Otolaryngology, Massachusetts Eye and Ear, Boston, Massachusetts, U.S.A.

Article Synopsis
  • The study aimed to evaluate the effects of hypoglossal nerve stimulation (HGNS) and expansion pharyngoplasty (EP) on patients with obstructive sleep apnea (OSA) to understand potential respiratory complications post-surgery.
  • Thirty patients (19 received HGNS and 11 received EP) were monitored using the NightOwl Mini device for changes in the apnea-hypopnea index (AHI) and oxygen levels after surgery.
  • Results showed no significant changes in AHI after HGNS, indicating no need for extended overnight observation, whereas EP led to increased AHI and hypoxemia, suggesting tailored postoperative care is necessary for those patients.

Role of Paraoxonase 2 in Airway Epithelial Response to Oxidant Stress.

Antioxidants (Basel)

Asthma is a widespread chronic lung disease characterized by airway inflammation and hyperresponsiveness. This airway inflammation is classified by either the presence (T2-high) or absence (T2-low) of high levels of eosinophils. Because most therapies for asthma target eosinophils and related pathways, treatment options for T2-low disease are limited.

Coherent reception, along with time- and wavelength-division multiplexing (TWDM), is a promising concept to simultaneously support multiple services in future high-speed point-to-multipoint passive optical networks (PONs). The next-generation PON 2 (NG-PON2) standard describes a TWDM-PON based on IM/DD intensity modulation and direct detection (IM/DD) which employs tunable-lasers and optical filters such as tunable optical filters or cyclic arrayed-waveguide gratings. Here, we investigate what we believe to be a novel coherent TWDM-PON architecture based on a frequency comb source in the optical line terminal (OLT), and thermally-tuned distributed-feedback (DFB) lasers in the optical network units (ONUs).
