Unusual paralytic shellfish poisoning toxins in Cayuga Lake, New York.

Toxicon

December 2024

USDA Agricultural Research Service, Hydrology and Remote Sensing Laboratory, 10300 Baltimore Ave, Beltsville, MD, 20705, USA. Electronic address:

Cyanobacteria blooms were detected in 2017 and 2018 in Cayuga Lake, New York, with paralytic shellfish poisoning toxins (PSPTs) the primary toxin detected. Analysis of these samples by HPLC with chemical oxidation, receptor binding assay, ELISA, and LC-MS/MS confirmed the presence of PSPTs but each method gave highly unusual results based on the theory establishing each method. The structures of the toxins could not be identified and may be 'unusual' or unelucidated PSPTs.

Aptamer-Encapsulated Cellular Nanoparticles for Neurotoxin Neutralization.

Adv Healthc Mater

January 2025

Aiiso Yufeng Li Family Department of Chemical and Nano Engineering, Shu and K.C. Chien and Peter Farrell Collaboratory, University of California San Diego, La Jolla, CA, 92093, USA.

Aptamers are single-stranded oligonucleotides that fold into defined architectures for specific target binding. In this study, aptamers are selected that specifically bind to small-molecule neurotoxins and encapsulate them into cell membrane-coated nanoparticles (referred to as 'cellular nanoparticles' or 'CNPs') for effective neutralization of neurotoxins. Specifically, six different aptamers are selected that bind to saxitoxin (STX) or tetrodotoxin (TTX) and encapsulate them into metal-organic framework cores, which are then coated with neuronal cell membrane.

Article Synopsis
  • The study examines the relationship between the characteristics of pufferfish PSTBP proteoforms and their thermal stability across four Takifugu species.
  • The researchers used methods like Western blot analysis and LC-MS/MS to confirm the heat-tolerance of these proteins, especially in T. rubripes.
  • Findings indicate that heat-stable PSTBP proteoforms are genetically conserved within the genus, enhancing our understanding of toxin transmission in seafood and its associated risks.

Saxitoxin (STX), an exceptionally potent marine toxin for which no antidote is currently available, is produced by methanogens and cyanobacteria. This poses a significant threat to both shellfish aquaculture and human health. Consequently, the development of a rapid, highly sensitive STX detection method is of great significance.

Development of a Multiplexed Electrochemical Aptasensor for the Detection of Cyanotoxins.

Biosensors (Basel)

May 2024

Department of Chemistry, Alfaisal University, Al Zahrawi Street, Al Maather, Al Takhassusi Road, Riyadh 11533, Saudi Arabia.

In this study, we report a multiplexed platform for the simultaneous determination of five marine toxins. The proposed biosensor is based on a disposable electrical printed (DEP) microarray composed of eight individually addressable carbon electrodes. The electrodeposition of gold nanoparticles on the carbon surface offers high conductivity and enlarges the electroactive area.
