The global outbreak and rapid spread of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have created an urgent need for large-scale testing of populations. There is a demand for high-throughput testing protocols that can be used for efficient and rapid testing of clinical specimens. We evaluated a pooled PCR protocol for testing nasopharyngeal (NP) swabs using known positive/negative and untested clinical samples that were assigned to pools of 5 or 10. In total, 630 samples were used in this study. Individual positive samples with cycle threshold ( ) values as high as 33 could be consistently detected when pooled with 4 negative samples (pool of 5), and individual positive samples with values up to 31 could be consistently detected when pooled with 9 negative samples (pool of 10). Pooling of up to 5 samples can be employed in laboratories for the diagnosis of COVID-19 for efficient utilization of resources, rapid screening of a greater number of people, and faster reporting of test results.

Introduction: The sequential parallel comparison design has emerged as a valuable tool in clinical trials with high placebo response rates. To further enhance its efficiency and effectiveness, adaptive strategies, such as sample size adjustment and allocation ratio modification can be employed.

Methods: We compared the performance of Jennison and Turnbull's method and the Promising Zone approach for sample size adjustment in a two-phase sequential parallel comparison design study.

The present study examined the effects of cultural factors(ethnic identity, acculturation, perceived discrimination, and religiosity), derived from the Multicultural Assessment-Intervention Process (MAIP) model, on attitudes toward prescription drug use among Iranian/Persian Americans across the United States. The study consisted of a final sample of 454 Iranian/Persian American adult participants. The results indicated that Iranian/Persian American attitudes toward prescription drug use are impacted by demographic and cultural factors.

Background: This study investigates the role of small ubiquitin-like modifier (SUMO)-specific peptidase 5 (SENP5), a key regulator of SUMOylation, in esophageal squamous cell carcinoma (ESCC), a lethal disease, and its underlying molecular mechanisms.

Methods: Differentially expressed genes between ESCC mouse oesophageal cancer tissues and normal tissues were analysed via RNA-seq; among them, SENP5 expression was upregulated, and this gene was selected for further analysis. Immunohistochemistry and western blotting were then used to validate the increased protein level of SENP5 in both mouse and human ESCC samples.

Background: This study investigated relationships between low-income adolescent drinkers' frequent alcohol use and five factors: social disorganization, social structural, social integration, mental health, and access to healthcare.

Objective: A sample of 1,256 low-income adolescent drinkers and caregivers were extracted from the Future of Families and Child Wellbeing Study.

Results: Logistic regression yielded results showing adolescent drinkers' weekly drinking to be associated positively with Hispanic adolescents, drinking peers, adolescents' depression/anxiety, and caregiver's daily drinking.

Research evidence has demonstrated a significant association between hypertrophic cardiomyopathy (HCM) and atrial fibrillation (AF), but the causality and pattern of this link remain unexplored. Therefore, this study investigated the causal relationship between HCM and AF using a two-sample and bidirectional Mendelian randomization (MR) approach. Additionally, this assessed the role of cardiovascular proteins (CPs) associated with cardiovascular diseases between HCM and AF by applying a two-step MR analysis.
