Background: Paromomycin is a 2-deoxystreptamine aminocyclitol aminoglycoside antibiotic with broad spectrum activity against Gram-negative, Gram-positive bacteria and many protozoa. This study introduces a strategy for paromomycin production through solid-state fermentation using Streptomyces rimosus subsp. paromomycinus NRRL 2455. Solid state fermentation has gained enormous attention in the development of several products because of their numerous advantages over submerged liquid fermentation. After selecting the best solid substrate, a time course study of paromomycin production was carried out followed by optimization of environmental conditions using response surface methodology. Paromomycin yields obtained using this technique were also compared to those obtained using submerged liquid fermentation.
Results: Upon screening of 6 different substrates, maximum paromomycin concentration (0.51 mg/g initial dry solids) was obtained with the cost-effective agro-industrial byproduct, corn bran, impregnated with aminoglycoside production media. Optimization of environmental conditions using D-optimal design yielded a 4.3-fold enhancement in paromomycin concentration reaching 2.21 mg/g initial dry solids at a pH of 8.5, inoculum size of 5% v/w and a temperature of 30 °C.
Conclusion: Compared to submerged liquid fermentation, solid state fermentation resulted in comparable paromomycin concentrations, cost reduction of raw materials, less energy consumption and waste water discharge, which have major implications in industrial fermentation. Therefore, solid state fermentation is a promising alternative to submerged liquid fermentation for paromomycin production. To the best of our knowledge, this is the first report on the optimized paromomycin production through solid state fermentation process.

Pillar stability has garnered significant attention owing to the effects of pillars on coal resource recovery rate, coal pillar stability, and coal bump risk. This study examined the roadway stability control principles of conventional and yield coal pillars. The conventional coal pillars were designed as load-bearing structures with a high load-bearing capacity to carry most of the abutment load, while yield coal pillars were designed as buffer structures for transferring rapidly increasing abutment loads to adjacent solid coal ribs by progressive deformation.

We report the synthesis, crystal structure, and magnetic properties of a new Kitaev honeycomb cobaltate, KCoAsO, which crystallizes in two distinct forms: P2/c and R[Formula: see text] space groups. Magnetic measurements reveal ordering temperatures of ~ 14 K for the P2/c structure and ~ 10.5 K for the R[Formula: see text] structure.

The development of acid-stable water oxidation electrocatalysts is crucial for high-performance energy conversion devices. Different from traditional nanostructuring, here we employ an innovative microwave-mediated electron-phonon coupling technique to assemble specific Ru atomic patterns (instead of random Ru-particle depositions) on MnCrO surfaces (Ru-MnCrO) in RuCl solution because hydrated Ru-ion complexes can be uniformly activated to replace some Mn sites at nearby Cr-dopants through microwave-triggered energy coherent superposition with molecular rotations and collisions. This selective rearrangement in Ru-MnCrO with particular spin-differentiated polarizations can induce localized spin domain inversion from reversed to parallel direction, which makes Ru-MnCrO demonstrate a high current density of 1.

Adoptive transfer of genetically or nanoparticle-engineered macrophages represents a promising cell therapy modality for treatment of solid tumor. However, the therapeutic efficacy is suboptimal without achieving a complete tumor regression, and the underlying mechanism remains elusive. Here, we discover a subpopulation of cancer cells with upregulated CD133 and programmed death-ligand 1 in mouse melanoma, resistant to the phagocytosis by the transferred macrophages.

Background: When a family decides to donate the organs or tissues of their child with brain death, it is necessary to consider which organs or tissues will be donated. This phenomenon presents an ethical dilemma that is underexplored in the scientific literature, making it essential to examine this context to understand how refusals occur within donations.
Objective: To analyze the rates and trends of specific refusals for each organ and tissue from pediatric donors with brain death occurring between 2001 and 2020 in an Organ Procurement Organization in the State of São Paulo, Brazil.
