DC. (CH) is found in many countries in Southeast Asia. This plant has been reported for anti-microbial, anti-cancer and anti-inflammatory bioactivities. However, the anti-inflammatory and anti-inflammasome properties of the leaves remain poorly understood. This study aimed to investigate the effect of CH leaves on NLRP3 and NF-κB signaling pathways. CH leaves were sequentially extracted using hexane, ethyl acetate and 95% ethanol to give three crude extracts. An active compound, lupeol was fractionated from the ethanolic extract using chromatographic techniques, and its structure was identified and confirmed by spectroscopic methods. Anti-inflammatory activities were observed on both lipopolysaccharide-stimulated and NLRP3 adenosine triphosphate-induced macrophages. The release of pro-inflammatory cytokines (IL-1β, IL-6 and TNF-α) was analyzed by Enzyme-Linked Immunosorbent Assay (ELISA). Real-time qRT-polymerase chain reaction (PCR) was used to measure inflammatory-associated gene expression. NF-κB protein expressions were investigated using the immunoblotting technique. The active fraction of ethanolic CH leaves and lupeol significantly reduced the release of pro-inflammatory cytokines and suppressed the expression of both inflammasome genes and NF-κB proteins. The ethanolic extract of CH leaves and lupeol showed potent anti-inflammatory activities by targeting NF-κB and NLRP3 signaling pathways.

Background: X-ray grating-based dark-field imaging can sense the small angle scattering caused by object's micro-structures. This technique is sensitive to the porous microstructure of lung alveoli and has the potential to detect lung diseases at an early stage. Up to now, a human-scale dark-field CT (DF-CT) prototype has been built for lung imaging.

With enrichment of tetracycline (TC) in ecosystems, its accurate detection has become a major concern. Noble-metal nano-particles have attracted great interest as potential materials for sensing applications because of their remarkable electrical properties and adaptability. Herein, a novel electro-chemical detection technique based on carbon nano-tubes (CNTs) as the support material is developed to detect TC with high precision.

The roles of mitochondria in global and local intracellular calcium signalling.

Nat Rev Mol Cell Biol

January 2025

MitoCare Center, Department of Pathology and Genomic Medicine, Thomas Jefferson University, Philadelphia, PA, USA.

Activation of Ca channels in Ca stores in organelles and the plasma membrane generates cytoplasmic calcium ([Ca]) signals that control almost every aspect of cell function, including metabolism, vesicle fusion and contraction. Mitochondria have a high capacity for Ca uptake and chelation, alongside efficient Ca release mechanisms. Still, mitochondria do not store Ca in a prolonged manner under physiological conditions and lack the capacity to generate global [Ca] signals.

Upon exposure to salt stress, calcium signaling in plants activates various stress-responsive genes and proteins along with enhancement in antioxidant defense to eventually regulate the cellular homeostasis for reducing cytosolic sodium levels. The coordination among the calcium signaling molecules and transporters plays a crucial role in salinity tolerance. In the present study, twenty-one diverse indigenous rice genotypes were evaluated for salt tolerance during the early seedling stage, and out of that nine genotypes were further selected for physio-biochemical study.

Department of Rheumatology and Immunology, Jingmen People's Hospital, JingChu University of Technology Affiliated Jingmen People's Hospital, No.39 Xiangshan Road Dongbao Zone, Jingmen, 448000, China.

Breast invasive carcinoma (BRCA) affects women worldwide, and despite advancements in diagnosis, prevention, and treatment, outcomes remain suboptimal. TNIP1, a novel target involved in multiple immune signaling pathways, influences tumor development and survival. However, the connection between BRCA and TNIP1 remains unclear.
