A novel pH-sensitive hydrogel system consisting of poly(methacrylic acid--ethylene glycol) (P(MAA--EG)) and acryloyl group modified-cholesterol-bearing pullulan (CHPOA) nanogels was developed for the controlled delivery of an anticonvulsant drug, pregabalin (PGB). Here, the hydrophilic hydrogel network provides the pH-sensitive swelling behavior, whereas nanogel components form separate reservoirs for the delivery of drugs with different hydrophobicities. These nanocarrier-integrated hybrid gels were synthesized through both surface-initiated and bulk photopolymerization approaches. The swelling and drug release behavior of these pH-responsive hydrogels synthesized by different photopolymerization approaches at visible and UV light wavelenghts were studied at acidic and basic pH values. Nanogel-integrated hydrogels exhibited higher swelling behavior compared to plain hydrogels in reversible swelling experiments. Similarly, the presence of nanogels in hydrogel network enhanced the loading and release percentages of PGB and the release was analyzed to describe the mode of transport through the network. In vitro cytotoxicity assay suggests that hydrogels in altered groups are nontoxic. This is the first report about the visible light-induced synthesis of a pH-responsive network incorporated CHPOA nanogels. Responsive and multifunctional properties of this system could be used for pH-triggered release of therapeutic molecules for clinical applications.

The receptor activator of NF-κB ligand (RANKL)-binding peptide is known to accelerate bone morphogenetic protein (BMP)-2-induced bone formation. Cholesterol-bearing pullulan (CHP)-OA nanogel-crosslinked PEG gel (CHP-OA nanogel-hydrogel) was shown to release the RANKL-binding peptide sustainably; however, an appropriate scaffold for peptide-accelerated bone formation is not determined yet. This study compares the osteoconductivity of CHP-OA hydrogel and another CHP nanogel, CHP-A nanogel-crosslinked PEG gel (CHP-A nanogel-hydrogel), in the bone formation induced by BMP-2 and the peptide.

Cell-based therapies hold significant advantages in comparison with the traditional drug-based or injection-based treatments. However, for long-term functional cellular implants, immune acceptance must be established. To accomplish the acceptance of the implanted cells, various biomaterial systems have been studied.

Different approaches have been developed to improve the scaffold properties that provide structural support and biological interaction to achieve the desired environment for tissue regeneration. We previously reported that addition of human fibroblast growth factor 18 (hFGF18) to acryloyl group-modified cholesterol-bearing pullulan (CHPOA) nanogel-crosslinked (NanoClik) hydrogels that contain human bone morphogenetic protein 2 (hBMP2) stabilized bone healing in mouse calvarial defect model. In this study, we evaluated the use of disc-shaped dried nanogel-crosslinked gel as carriers of growth factors in order to seek possible clinical application in future.

Sequential Coating of Insulin Secreting Beta Cells within Multilayers of Polysaccharide Nanogels.

Pancreatic islet transplantation has emerged as a promising treatment for type-1 diabetes (T1D); however, its clinical application is still limited by the life-long use of immunosuppressive drugs, insufficient number of islets to achieve normoglycemia, and large transplantation volume. This paper reports a unique approach for nanothin coating of insulin secreting beta cell aggregates. The coating is based on hydrophobic and covalent interactions between natural acrylate modified cholesterol bearing pullulan (CHPOA) nanogels and MIN6 beta cell aggregates.
