Purpose: Dynamic contrast-enhanced MRI (DCE-MRI) represents the only available approach for glymphatic cerebrospinal fluid (CSF) flow 3D mapping in the brain of living animals and humans. The purpose of this study was to develop a novel DCE-MRI protocol for mapping of the glymphatic system transport with improved spatiotemporal resolution, and to validate the new protocol by comparing the transport in mice anesthetized with either isoflurane or ketamine/xylazine.
Methods: The contrast agent, gadobutrol, was administered into the CSF of the cisterna magna and its transport visualized continuously on a 9.4T preclinical scanner using 3D fast-imaging with a steady-state free-precession sequence (3D-FISP), which has a spatial resolution of 0.001 mm and a temporal resolution of 30 s. The MR signals were measured dynamically for 60 min in multiple volumes of interest covering the entire CSF space and brain parenchyma.
Results: The results confirm earlier findings that glymphatic CSF influx is higher under ketamine/xylazine than with isoflurane anesthesia. This was extended to account for new details about the distinct CSF efflux pathways under the two anesthetic regimens. Dynamic contrast MR shows that CSF clearance occurs mainly along the vagus nerve near the jugular vein under isoflurane and via the olfactory bulb under ketamine/xylazine.
Conclusion: The improved spatial and temporal sampling rates afforded by 3D-FISP shed new light on the pharmacological modulation of CSF efflux paths. The present observations may have the potential to set a new standard for future experimental DCE-MRI studies of the glymphatic system.

Corticospinal motor neurons (CSMN), located in the motor cortex of the brain, are one of the key components of the motor neuron circuitry. They are in part responsible for the initiation and modulation of voluntary movement, and their degeneration is the hallmark for numerous diseases, such as amyotrophic lateral sclerosis (ALS), hereditary spastic paraplegia, and primary lateral sclerosis. Cortical hyperexcitation followed by in-excitability suggests the early involvement of cortical dysfunction in ALS pathology.

Background: The evolution and development of flowers are biologically essential and of broad interest. Maize and sorghum have similar morphologies and phylogeny while harboring different inflorescence architecture. The difference in flower architecture between these two species is likely due to spatiotemporal gene expression regulation, and they are a good model for researching the evolution of flower development.

In recent decades, global climate change and rapid urbanization have aggravated the urban heat island (UHI) effect, affecting the well-being of urban citizens. Although this significant phenomenon is more pronounced in larger metropolitan areas due to extensive impervious surfaces, small- and medium-sized cities also experience UHI effects, yet research on UHI in these cities is rare, emphasizing the importance of land surface temperature (LST) as a key parameter for studying UHI dynamics. Therefore, this paper focuses on the evaluation of LST and land cover (LC) changes in the city of Prešov, Slovakia, a typical medium-sized European city that has recently undergone significant LC changes.

The visual environment of sign language users is markedly distinct in its spatiotemporal parameters compared to that of non-signers. Although the importance of temporal and spectral resolution in the auditory modality for language development is well established, the spectrotemporal parameters of visual attention necessary for sign language comprehension remain less understood. This study investigates visual temporal resolution in learners of American Sign Language (ASL) at various stages of acquisition to determine how experience with sign language affects perceptual sampling.

Background: The molecular etiology of tau-derived neurodegeneration remains poorly understood, reflected in the low success rate of clinical trials. Hence, aquiring a better understanding the molecular basis of tauopathies is a critical need.
Objective: To develop a versatile and reproducible system to study tau aggregation with high spatiotemporal control through optogenetics that will aid in investigating the differences in tau aggregation kinetics, the burden the burden of tau isoforms, and mutations and that will be suitable for high-throughput analysis of tauopathy-related mechanisms.
