Axonal degeneration is responsible for disease progression and accumulation of disability in many neurodegenerative conditions. The axonal degenerative process can generate a metastable pool of damaged axons that remain structurally and functionally viable but fated to degenerate in the absence of external intervention. SARM1, an NADase that depletes axonal energy stores upon activation, is the central driver of an evolutionarily conserved program of axonal degeneration. We identify a potent and selective small molecule isoquinoline inhibitor of SARM1 NADase that recapitulates the SARM1 phenotype and protects axons from degeneration induced by axotomy or mitochondrial dysfunction. SARM1 inhibition post-mitochondrial injury with rotenone allows recovery and rescues axons that already entered the metastable state. We conclude that SARM1 inhibition with small molecules has the potential to treat axonopathies of the central and peripheral nervous systems by preventing axonal degeneration and by allowing functional recovery of a metastable pool of damaged, but viable, axons.

The effect of temperature on the localized corrosion resistance and passive film characteristics of laser powder-bed fusion (LPBF) 316L (UNS S31603) was studied in a buffered 3.5 wt% NaCl solution at 25, 50, and 75 °C. DC techniques such as cyclic potentiodynamic polarization showed lower passive current densities, high breakdown potentials, and a higher resistance to initial breakdown compared with wrought 316L samples at all temperatures.

Metastable materials are abundant in nature and technology, showcasing remarkable properties that inspire innovative materials design. However, traditional crystal structure prediction methods, which rely solely on energetic factors to determine a structure's fitness, are not suitable for predicting the vast number of potentially synthesizable phases that represent a local minimum corresponding to a state in thermodynamic equilibrium. Here, we present a new approach for the prediction of metastable phases with specific structural features and interface this method with the XtalOpt evolutionary algorithm.

Numerical Simulation of Temperature Fields during Laser Welding-Brazing of Al/Ti Plates.

The formation of dissimilar weld joints, including Al/Ti joints, is an area of research supported by the need for weight reduction and corrosion resistance in automotive, aircraft, aeronautic, and other industries. Depending on the cooling rates and chemical composition, rapid solidification of Al/Ti alloys during laser welding can lead to the development of different metastable phases and the formation of brittle intermetallic compounds (IMCs). The effort to successfully join aluminum to titanium alloys is associated with demands to minimize the thickness of brittle IMC zones by selecting appropriate welding parameters or applying suitable filler materials.

Heterogeneous levels of delta-like 4 within a multinucleated niche cell maintains muscle stem cell diversity.

The quiescent muscle stem cell (QSC) pool is heterogeneous and generally characterized by the presence and levels of intrinsic myogenic transcription factors. Whether extrinsic factors maintain the diversity of states across the QSC pool remains unknown. The muscle fiber is a multinucleated syncytium that serves as a niche to QSCs, raising the possibility that the muscle fiber regulates the diversity of states across the QSC pool.

The samples of the Al-15Fe (mass%) binary alloy that were additively manufactured by laser powder bed fusion (L-PBF) were exposed to intermediate temperatures (300 and 500 °C), and the thermally induced variations in their microstructural characteristics were investigated. The L-PBF-manufactured sample was found to have a microstructure comprising a stable θ-AlFe phase localized around melt-pool boundaries and several spherical metastable AlFe-phase particles surrounded by a nanoscale α-Al/AlFe cellular structure in the melt pools. The morphology of the θ phase remained almost unchanged even after 1000 h of exposure at 300 °C.
