RAC1 activity is critical for intestinal homeostasis, and is required for hyperproliferation driven by loss of the tumour suppressor gene Apc in the murine intestine. To avoid the impact of direct targeting upon homeostasis, we reasoned that indirect targeting of RAC1 via RAC-GEFs might be effective. Transcriptional profiling of Apc deficient intestinal tissue identified Vav3 and Tiam1 as key targets. Deletion of these indicated that while TIAM1 deficiency could suppress Apc-driven hyperproliferation, it had no impact upon tumourigenesis, while VAV3 deficiency had no effect. Intriguingly, deletion of either gene resulted in upregulation of Vav2, with subsequent targeting of all three (Vav2 Vav3 Tiam1), profoundly suppressing hyperproliferation, tumourigenesis and RAC1 activity, without impacting normal homeostasis. Critically, the observed RAC-GEF dependency was negated by oncogenic KRAS mutation. Together, these data demonstrate that while targeting RAC-GEF molecules may have therapeutic impact at early stages, this benefit may be lost in late stage disease.

Pancreatic ductal adenocarcinoma carries a dismal prognosis, with high rates of metastasis and few treatment options. Hyperactivation of KRAS in almost all tumours drives RAC1 activation, conferring enhanced migratory and proliferative capacity as well as macropinocytosis. Macropinocytosis is well understood as a nutrient scavenging mechanism, but little is known about its functions in trafficking of signalling receptors.

Integrated analysis of tumour-derived exosome-related immune genes to predict progression and immune status of hepatocellular carcinoma.

Tumour-derived exosomes (TDEs) play an important role in tumourigenesis and progression by regulating components in the tumour microenvironment (TME), however, the role of TDE-related immune genes in hepatocellular carcinoma is not fully known. We systematically analysed TDE genes from ExoCarta and immune genes from Immport,Machine learning ultimately identified eight TDE-related prognostic immune genes and used them as the basis for constructing a risk model, which was constructed to better predict patients with hepatocellular carcinoma (HCC) compared with published prognostic models. There were significant differences between the high and low risk groups in terms of biological functioning.

DAB2IP attenuates chemoresistance of triple-negative breast cancer through sequestration of RAC1 to prevent β-catenin nuclear accumulation.

Clin Transl Med

December 2022

Department of Breast Oncology, Sun Yat-sen University Cancer Center, State Key Laboratory of Oncology in South China, Collaborative Innovation Center of Cancer Medicine, Guangzhou, China.

Background: Although chemotherapy, the most widely used systemic treatment in triple-negative breast cancer (TNBC), markedly improved the patients' outcome, chemoresistance always occurs. This study purposed to explore new therapeutic strategies for the treatment of chemoresistance.

Methods And Results: The expression and prognostic value of DAB2IP were investigated in TNBC tissues and cell lines.

Article Synopsis
  • Recent findings show that mutations in the VAV1 gene are linked to peripheral T cell lymphoma and non-small-cell lung cancer (NSCLC).
  • Researchers created a gene-edited mouse model to study the impact of a specific VAV1 mutation, which did not cause cancer alone but led to T cell lymphoma when combined with the loss of the Trp53 gene.
  • The study revealed that VAV1 mutations influence tumor development in specific cell types and interact with other mutations, highlighting the complexity of cancer mechanisms.

Negative regulation of TGFβ-induced apoptosis by RAC1B enhances intestinal tumourigenesis.

Article Synopsis
  • * In a study with mice that have colorectal cancer, they found that high levels of RAC1B didn't help tumors spread but instead made them form faster and hurt the mice's chances of living longer.
  • * The study showed that tumors with more RAC1B avoided cell death and had weak signals from a pathway called TGFβ, which usually helps control cell growth and death.
