Consumption of illicit pharmaceutical products containing sibutramine has been reported to cause cardiovascular toxicity problems. This study aimed to demonstrate the toxicity profile of sibutramine, and thereby provide important implications for the development of more effective strategies in both clinical approaches and drug design studies. Action potentials (APs) were determined from freshly isolated ventricular cardiomyocytes with whole-cell configuration of current clamp as online. The maximum amplitude of APs (MAPs), the resting membrane potential (RMP), and AP duration from the repolarization phases were calculated from original records. The voltage-dependent K-channel currents (I) were recorded in the presence of external Cd and both inward and outward parts of the current were calculated, while their expression levels were determined with qPCR. The levels of intracellular free Ca and H (pH) as well as reactive oxygen species (ROS) were measured using either a ratiometric micro-spectrofluorometer or confocal microscope. The mechanical activity of isolated hearts was observed with Langendorff-perfusion system. Acute sibutramine applications (10-10 M) induced significant alterations in both MAPs and RMP as well as the repolarization phases of APs and I in a concentration-dependent manner. Sibutramine (10 μM) induced Ca-release from the sarcoplasmic reticulum under either electrical or caffeine stimulation, whereas it depressed left ventricular developed pressure with a marked decrease in the end-diastolic pressure. pH inhibition by sibutramine supports the observed negative alterations in contractility. Changes in mRNA levels of different I subunits are consistent with the acute inhibition of the repolarizing I, affecting AP parameters, and provoke the cardiotoxicity.

Background: The aim of this simulation was to examine the utility of a novel ECG-based index of cardiac action potential (AP) triangulation, the Tstart-to-Tpeak (TsTp) interval-to-JTstart (JTs) interval ratio, for assessment of changes in AP profile imposed through variations in the duration of the plateau phase and the phase 3 repolarization.
Methods: ECGs were simulated using a realistic rabbit model based on experimental data. The AP plateau was measured at APD30, and the phase 3 was assessed as APD90-to-APD30 difference (AP durations at 90 % and 30 % repolarization, respectively).

This research assessed blood pressure and dispersion mapping in reliance on age (Myocardium and Rhythm indices), the incidence of their deviations from the standard values in comparison with the tendency in mortality from diseases of the blood circulation system. For this purpose, we studied test results of 602 men and 776 women (total 1 378 people) aged 17-90 years who permanently inhabited Magadan city and underwent medical examination in 2016-2019, and mortality rates from circulatory system diseases during this period were estimated. The research showed a strong link of the increasing age to the growth in the blood pressure variables, with hypertensive focus, and the deviations in the ECG dispersion characteristics of the Myocardium and Rhythm indices tended to grow bigger.

The waveform of ventricular action potential (AP) is a key determinant of the cardiac cycle, a marker of beating pathophysiology, and a target for anti-arrhythmic drug design. The information contained in the waveform, though, is limited to the actual dynamics of the AP under consideration. By measuring quasi-instantaneous current-voltage relationships during repolarization, I propose a three-dimensional representation of the ventricular AP which includes potential dynamic responses that the beat can show when electrically perturbed.
