Early success with brentuximab vedotin in treating classical Hodgkin lymphoma spurred an influx of at least 20 monomethyl auristatin E (MMAE) antibody-drug conjugates (ADCs) into clinical trials. While three MMAE-ADCs have been approved, most of these conjugates are no longer being investigated in clinical trials. Some auristatin conjugates show limited or no efficacy at tolerated doses, but even for drugs driving initial remissions, tumor regrowth and metastasis often rapidly occur. Here we describe the development of second-generation therapeutic ADCs targeting Lymphocyte antigen 6E (Ly6E) where the tubulin polymerization inhibitor MMAE (Compound ) is replaced with DNA-damaging agents intended to drive increased durability of response. Comparison of a -cyclopropyl benzoindol-4-one (CBI)-dimer (compound ) to MMAE showed increased potency, activity across more cell lines, and resistance to efflux by -glycoprotein, a drug transporter commonly upregulated in tumors. Both anti-Ly6E-CBI and -MMAE conjugates drove single-dose efficacy in xenograft and patient-derived xenograft models, but CBI-dimer conjugates showed reduced tumor outgrowth following multiple weeks of treatment, suggesting that they are less susceptible to developing resistance. In parallel, we explored approaches to optimize the targeting antibody. In contrast to immunization with recombinant Ly6E or Ly6E DNA, immunization with virus-like particles generated a high-affinity anti-Ly6E antibody. Conjugates to this antibody improve efficacy versus a previous clinical candidate both and with multiple cytotoxics. Conjugation of compound to the second-generation antibody results in a substantially improved ADC with promising preclinical efficacy.

Benralizumab is an anti-IL-5 receptor alpha monoclonal antibody that induces the near-complete depletion of eosinophils. This study aimed to evaluate the long-term safety and effectiveness of benralizumab in patients with severe eosinophilic asthma (SEA) over an extended 48-month follow-up period, offering one of the longest real-world perspectives available. This was a single-arm, retrospective, observational, multicenter study involving 123 SEA patients treated with benralizumab at a dosage of 30 mg every 4 weeks for the first 3 doses and then every 8 weeks.

Hydraulic structures are frequently subjected to soft-water or acidic environments, necessitating serious consideration of the long-term effects of calcium leaching on the durability of concrete structures. Three types of common Portland cement (ordinary Portland cement, moderate-heat cement, and low-heat cement) paste samples widely applied to hydraulic concrete were immersed in a 6 mol/L NHCl solution to simulate accelerated calcium leaching behavior. The increasing challenges related to the reliability and durability of steel pipeline infrastructure necessitate a detailed understanding of degradation and failure mechanisms. This study focuses on selective corrosion and erosion as critical factors, analyzing their impact on pipeline integrity using advanced methods, including macroscopic analysis, corrosion testing, microscopic examination, tensile strength testing, and finite element method (FEM) modeling. Selective corrosion in the heat-affected zones (HAZs) of longitudinal welds was identified as the dominant degradation mechanism, with pit depths reaching up to 6 mm, leading to tensile strength reductions of 30%.

We investigated the phase transitions, mechanical properties, and chemical durability of a composition of 9 mol% CaO-stabilized zirconia (9CSZ) doped with 2-4 mol% CeO under thermal stress against molten slag.
