Isolation and characterization of cultured chicken oviduct epithelial cells and in vitro validation of constructed ovalbumin promoter in these cells. | LitMetric

AI Article Synopsis

Article Abstract

Objective: Transgenic hens hold a great promise to produce various valuable proteins. Through virus transduction into stage X embryo, the transgene expression under the control of constructed chicken ovalbumin promoters has been successfully achieved. However, a validation system that can evaluate differently developed ovalbumin promoters in in vitro, remains to be developed.

Methods: In the present study, chicken oviduct epithelial cells (cOECs) were isolated from oviduct tissue and shortly cultured with keratinocyte complete medium supplemented with chicken serum. The isolated cells were characterized with immunofluorescence, western blot, and flow cytometry using oviduct-specific marker. Chicken mutated ovalbumin promoter (Mut-4.4-kb-pOV) was validated in these cells using luciferase reporter analysis.

Results: The isolated cOECs revealed that the oviduct-specific marker, ovalbumin protein, was clearly detected by immunofluorescence, western blot, and flow cytometry analysis revealed that approximately 79.40% of the cells contained this protein. Also, luciferase reporter analysis showed that the constructed Mut-4.4-kb-pOV exhibited 7.1-fold (p<0.001) higher activity in the cOECs.

Conclusion: Collectively, these results demonstrate the efficient isolation and characterization of cOECs and validate the activity of the constructed ovalbumin promoter in the cultured cOECs. The in vitro validation of the recombinant promoter activity in cOECs can facilitate the production of efficient transgenic chickens for potential use as bioreactors.

Background: The gut, the ureter, or the Fallopian tube all transport biological fluids by generating trains of propagating smooth muscle constrictions collectively known as peristalsis. These tubes connect body compartments at different pressures. We extend here Poiseuille's experiments on liquid flow in inert tubes to an active, mechanosensitive tube: the intestine.

Article Synopsis
  • - The study explored how different dietary ratios of linseed and soybean oils affected egg quality and health in 70-week-old Jinghong laying hens, mixing them into four groups for a 4-week period with varying oil ratios.
  • - No significant differences were found in egg production or overall feed intake among the groups, but the T2 group (2:1 linseed to soybean oil) exhibited higher follicle counts, and the T3 group (1:2 ratio) showed better albumen height and antioxidant enzyme activity compared to the T4 group (all soybean oil).
  • - The findings indicated that a 1:2 ratio of linseed to soybean oil improved egg quality by enhancing antioxidant capacity and reducing oxidative stress in the

Article Synopsis
  • This study provides the first detailed analysis of a new variant of infectious bronchitis virus (IBV) called D181 found in poultry in Morocco, focusing on both its pathological and molecular characteristics.
  • The research involved six poultry farms with various age groups, revealing severe egg production issues in layers and respiratory problems in broilers, alongside significant necropsy findings like hemorrhagic ovarian follicles and respiratory distress.
  • Molecular analysis showed a close genetic relationship between the Moroccan D181 strains and similar strains identified in the Netherlands in 2018, suggesting potential pathways for disease transmission.

  • Salmonella Reading (S. Reading) has emerged as a significant foodborne pathogen causing outbreaks linked to contaminated turkey products in North America, making understanding its transmission critical for prevention.
  • This study involved infecting 32 broiler breeder hens with two different bioluminescent strains of S. Reading to investigate their ability to colonize the hens' reproductive tissues and contaminate eggs.
  • Results indicated that the outbreak strain contaminated a higher percentage of eggshells compared to the nonoutbreak strain, suggesting S. Reading can effectively colonize and pose risks for egg contamination, with further research needed on its viability during incubation.

Article Synopsis
  • Intermittent fertilization intensity (IFI) in chickens affects fertilization rates, but its genetic basis remains unclear; this study investigates that relationship using 939 Wenchang chickens.
  • Through genome-wide association studies, researchers identified 35 significant SNP variants linked to IFI on chromosome 8, particularly in the intron of a gene homogenous with human LMO4, suggesting a shared regulatory mechanism.
  • The study highlights the role of immune regulation in the oviduct, with LMO4's activation related to inflammation, indicating that modifying LOC101750715 and LMO4 could lead to better fertilization outcomes in chickens.
