Article Abstract

Background: Thrombospondin-related anonymous protein (TRAP) has been described as a potential vaccine candidate for several diseases caused by apicomplexan parasites. However, this protein and members of this family have not yet been characterized in Babesia bigemina, one of the most prevalent species causing bovine babesiosis.

Methods: The 3186-bp Babesia bigemina TRAP-1 (BbiTRAP-1) gene was identified by a bioinformatics search using the B. bovis TRAP-1 sequence. Members of the TRAP and TRAP-related protein families (TRP) were identified in Babesia and Theileria through a search of the TSP-1 adhesive domain, which is the hallmark motif in both proteins. Structural modeling and phylogenetic analysis were performed with the identified TRAP proteins. A truncated recombinant BbiTRAP-1 that migrates at approximately 107 kDa and specific antisera were produced and used in Western blot analysis and indirect fluorescent antibody tests (IFAT). B-cell epitopes with neutralizing activity in BbiTRAP-1 were defined by enzyme-linked immunosorbent assays (ELISA) and invasion assays.

Results: Three members of the TRAP family of proteins were identified in B. bigemina (BbiTRAP-1 to -3). All are type 1 transmembrane proteins containing the von Willebrand factor A (vWFA), thrombospondin type 1 (TSP-1), and cytoplasmic C-terminus domains, as well as transmembrane regions. The BbiTRAP-1 predicted structure also contains a metal ion-dependent adhesion site for interaction with the host cell. The TRP family in Babesia and Theileria species contains the canonical TSP-1 domain but lacks the vWFA domain and together with TRAP define a novel gene superfamily. A variable number of tandem repeat units are present in BbiTRAP-1 and could be used for strain genotyping. Western blot and IFAT analysis confirmed the expression of BbiTRAP-1 by blood-stage parasites. Partial recognition by a panel of sera from B. bigemina-infected cattle in ELISAs using truncated BbiTRAP-1 suggests that this protein is not an immunodominant antigen. Additionally, bovine anti-recombinant BbiTRAP-1 antibodies were found to be capable of neutralizing merozoite invasion in vitro.

Conclusions: We have identified the TRAP and TRP gene families in several Babesia and Theileria species and characterized BbiTRAP-1 as a novel antigen of B. bigemina. The functional relevance and presence of neutralization-sensitive B-cell epitopes suggest that BbiTRAP-1 could be included in tests for future vaccine candidates against B. bigemina.

Introduction: , a parasite on the body surface of sheep, directly attacks the host through biting and sucking blood and may also transmit pathogens in the process. There are currently only a few studies on the microbial composition of , while there are no such studies on pupae.

Methods: In this study, samples AT-1 to AT-4 each contained four individuals, while sample AT-5 comprised four pupae, all used for metagenomic sequencing and analysis.

Background/objectives: Trypanosomiasis and bovine babesiosis correspond to important diseases that cause great economic losses, but there are still no studies evaluating their occurrence in herds of dairy cattle in Ceará. The aim of this study was to perform molecular diagnosis of the main hemoparasites of dairy cows in the microregion of the central hinterland of Ceará.

Methods: For the molecular diagnosis of parasites, genomic material was extracted and polymerase chain reaction directed to the cdCatL-like gene for and SS rRNA of and was performed.

Tick Infestation and Molecular Detection of Tick-Borne Pathogens from Indian Long-Eared Hedgehogs () in Pakistan.

Animals (Basel)

November 2024

Animal Sciences Division, Pakistan Agricultural Research Council, Islamabad 44000, Pakistan.

Hedgehogs can act as reservoirs for the transmission of tick-borne pathogens (TBPs) to domestic livestock, wild animals, and humans. Understanding host-tick dynamics is essential to evaluate the impact of TBPs. This study was conducted in Pakistan and aimed to determine the prevalence and species of TBPs in the blood and ticks of Indian long-eared hedgehogs captured from various environments.

is a major cause of bovine babesiosis, an economically devastating tick-borne disease that needs timely diagnosis for precise treatment. In present investigation, the detection efficacy of real-time PCR (qPCR) in comparison to conventional PCR and microscopy targeting 18 S ribosomal gene was evaluated on 95 bovines (70 cattle and 25 buffaloes) suspected for babesiosis. Real-time PCR was standardized with the 10-fold serial dilutions in duplication of the given positive control (2 × 10 copy number) ranging from 10 to 10 copy number/µL and mean Ct value of each dilution was taken to extrapolate the curve.

A 50-year-old question: Can imidocarb chemoprophylaxis ensure seroconversion for babesiosis in cattle under field conditions?

Vet Parasitol

Retired researcher at Instituto de Pesquisas Veterinárias Desidério Finamor (IPVDF), Eldorado do Sul, RS 92990-000, Brazil.

Article Synopsis
  • Bovine babesiosis, caused by Babesia bovis and Babesia bigemina, significantly impacts livestock and is often treated with imidocarb in areas lacking vaccines.
  • The study involved 15 heifers, with 12 receiving imidocarb and all exposed to ticks to evaluate the development of humoral immunity through seroconversion.
  • Results showed that after multiple treatments, a majority of the treated heifers did develop antibodies against the parasites, but not all animals seroconverted, highlighting potential limitations in the effectiveness of the chemoprophylaxis approach.
