Skeletal muscles are high-insulin tissues responsible for disposing of glucose via the highly regulated process of facilitated glucose transporter 4 (GLUT4). Impaired insulin action in diabetes, as well as disorders of GLUT4 vesicle trafficking in the muscle, are involved in defects in insulin-stimulated GLUT4 translocation. Since the Rab GTPases are the main regulators of vesicular membrane transport in exo- and endo-cytosis, in the present work, we studied the effect of olive leaf polyphenols (OLPs) on Rab8A, Rab13, and Rab14 proteins of the rat soleus muscle in a model of streptozotocin (SZT)-induced diabetes (DM) in a dose-dependent manner. Glucose, cholesterol, and triglyceride levels were determined in the blood, morphological changes of the muscle tissue were captured by hematoxylin and eosin histological staining, and expression of GLUT4, Rab8A, Rab13, and Rab14 proteins were analyzed in the rat soleus muscle by the immunofluorescence staining and immunoblotting. OLPs significantly reduced blood glucose level in all treated groups. Furthermore, significantly reduced blood triglycerides were found in the groups with the lowest and highest OLPs treatment. The dynamics of activation of Rab8A, Rab13, and Rab14 was OLPs dose-dependent and more effective at higher OLP doses. Thus, these results indicate a beneficial role of phenolic compounds from the olive leaf in the regulation of glucose homeostasis in the skeletal muscle.

Article Synopsis
  • Gestational Diabetes Mellitus (GDM) is a common pregnancy complication that can affect renal function and oxidative stress, prompting this study on the potential benefits of olive leaf extracts and stem cells.
  • Research involved pregnant rats with induced diabetes to measure various kidney and oxidative stress markers, finding that treatments significantly improved these conditions.
  • Results indicated that both treatments not only reduced harmful substance levels but also promoted cellular regeneration and improved kidney tissue structure, suggesting their effectiveness in managing GDM-related complications.

Castor bean (Ricinus communis) is cultivated agriculturally for oil and ornamentally for its bright foliage and seed. Ornamental castor bean has naturalized in many areas of the world, including the state of Sinaloa, Mexico, where it is not planted commercially. In a survey conducted in 2019 in Sinaloa, wild castor bean was found widely affected by a foliar blight with symptoms similar to Alternaria ricini previously described in the United States (Stevenson 1945) and in the state of Chiapas, Mexico (López-Guillén et al.

Fangfeng (Saposhnikovia divaricata) is a perennial plant belonging to the Umbelliferae family, and is widely cultivated as a traditional Chinese medicine plant used to treat various diseases in northern China. In August 2022, a widespread leaf spot disease emerged on the Fangfeng leaves across a 2.5-acre farmland located in the Naiman District of Tongliao City, China ( 44°17' N; 121°29' E), where 5,000 acres of Fangfeng had been cultivated.

Background/aims: Gestational Diabetes Mellitus (GDM), a prevalent complication in pregnancy, is characterized by the Diabetes Association as diabetes diagnosed in the second or third trimester, often remaining asymptomatic. This study investigates the intricate effects of Streptozotocin on pregnant rats, unraveling its impact on Gestational Type 2 Diabetes (GTD). The research delves into the potential therapeutic roles of mesenchymal stem cells (MSCs) and olive leaf extract (OLE) in mitigating the consequences of Streptozotocin-induced pancreatic impairment.
