Glass nanopipettes are widely used for various applications in nanosciences. In most of the applications, it is important to characterize their geometrical parameters, such as the aperture size and the inner cone angle at the tip region. For nanopipettes with sub-10 nm aperture and thin wall thickness, transmission electron microscopy (TEM) must be most instrumental in their precise geometrical measurement. However, this measurement has remained a challenge because heat generated by electron beam irradiation would largely deform sub-10 nm nanopipettes. Here, we provide methods for preparing TEM specimens that do not cause deformation of such tiny nanopipettes.

Plasma-activated Ringer's lactate solutions (PALs), which are Ringer's lactate solutions treated with non-thermal atmospheric-pressure plasma, have an anti-tumor effect and can be used for chemotherapy. As the anti-tumor effect of the PAL is influenced by the cell-treatment time, it is necessary to monitor the structural changes of the cell surface with non-invasive, nanoscale, and time-lapse imaging to understand the anti-tumor effect. In this study, to characterize the anti-tumor effect of the PAL, we used scanning ion conductance microscopy (SICM), using glass nanopipettes as probes, to visualize the structural changes of the cell surface.

Glass nano/micron pipettes, owing to their easy preparation, unique confined space at the tip, and modifiable inner surface of the tip, can capture the ion current signal caused by a single entity, making them widely used in the construction of highly sensitive and highly selective electrochemical sensors for single entity analysis. Compared with other solid-state nanopores, their conical nano-tip causes less damage to cells when inserted into them, thereby becoming a powerful tool for the analysis of important substances in cells. However, glass nanopipettes have some shortcomings, such as poor mechanical properties, difficulty in precise preparation (aperture less than 50 nm), and easy blockage during complex real sample detection, limiting their practicability.

Glass nanopipettes, as important sensing tools, have attracted great interest due to their wide range of applications in detecting single molecules, nanoparticles, and cells. In this study, we investigated the translocation behavior of nanorod particles through dual-nanopipettes using a transient continuum-based model based on an arbitrary Lagrangian-Eulerian approach. Our findings indicate that the translocation of nanorods is slowed down in the dual-nanopipette system, especially in the dual-nanopipette system with a nanobridge.

Cell-substrate interaction plays a critical role in determining the mechanical status of living cell membrane. Changes of substrate surface properties can significantly alter the cell mechanical microenvironment, leading to mechanical changes of cell membrane.
