Methods for simple and fast assembly of exchangeable standard DNA parts using Type II S restriction enzymes are becoming more and more popular in plant synthetic and molecular biology. These methods enable routine construction of large and complex multigene DNA structures. Two available frameworks emphasize either high cloning capacity (Modular Cloning, MoClo) or simplicity (GoldenBraid, GB). Here we present a set of novel α-level plasmids compatible with the GB convention that extend the ability of GB to rapidly assemble more complex genetic constructs, while maintaining compatibility with all existing GB parts as well as most MoClo parts and GB modules. With the use of our new plasmids, standard GB parts can be assembled into complex assemblies containing 1, 5, 10 and up to theoretically 50 units in each successive level of infinite loop assembly. Assembled DNA constructs can be also combined with conventional binary GB-assemblies (1, 2, 4, 8… units). We demonstrate the usefulness of our framework on single tube assembly of replicating plant expression constructs based on the geminivirus Bean yellow dwarf virus (BeYDV).

Due to their outstanding electrical and thermal properties, graphene and related materials have been proposed as ideal candidates for the development of lightweight systems for thermoelectric applications. Recently, the nanolaminate architecture that entails alternation of continuous graphene monolayers and ultrathin polymer films has been proposed as an efficient route for the development of composites with impressive physicochemical properties. In this work, we present a novel layer-by-layer approach for the fabrication of highly ordered, flexible, heat-resistant, and electrically conductive freestanding graphene/polymer nanolaminates through alternating Marangoni-driven self-assembly of reduced graphene oxide (rGO) and poly(ether imide) (PEI) films.

In this work, a series of three-dimensional (3D) SERS substrate were successfully fabricated by assembling silver nanoparticles (AgNPs) onto a porous gelatin sponge (GS) for highly sensitive thiram residues detection in vegetables. These 3D micro-nanostructures could induce the sufficient surface plasmon resonance (SPR) effect of noble metals on their surface and achieve high enrichment of pollutant molecules. As crystal violet (CV) was used as a probe molecule, the lowest CV solution could be detected at 10 M, and the enhancement factor (EF) was calculated to be 9.

One critical issue in hydrogen/deuterium exchange mass spectrometry (HDX MS) analysis is the deleterious back exchange. Herein, we report that when matrix-assisted laser desorption/ionization (MALDI) is used, the MALDI process itself can also cause significant back exchange. The back exchange occurred inside the reactive MALDI plume was investigated by depositing a fully deuterated sample prepared in DO on top of a preloaded dried layer of matrix.

Adenosin triphosphate (ATP)-dependent nucleosome remodeling factors sculpt the nucleosomal landscape of eukaryotic chromatin. They deposit, evict, or reposition nucleosomes along DNA in a process termed nucleosome sliding. Remodeling has traditionally been analyzed using mononucleosomes as a model substrate.

Swarm robots offer fascinating opportunities to perform complex tasks beyond the capabilities of individual machines. Just as a swarm of ants collectively moves large objects, similar functions can emerge within a group of robots through individual strategies based on local sensing. However, realizing collective functions with individually controlled microrobots is particularly challenging because of their micrometer size, large number of degrees of freedom, strong thermal noise relative to the propulsion speed, and complex physical coupling between neighboring microrobots.
