Activated leukocytes and the hemostatic system.

Activated leukocytes are capable of activating the blood-clotting system. Upon adequate stimulation (e.g., by endotoxin, by lectins, in the course of immune reactions, and in response to lymphokines of T cell origin), macrophages and monocytes synthesize tissue thromboplastin and expose it on their surface. Leukocyte proteases may interfere with blood coagulation by degrading clotting factors, in particular factors V, VIII, and XIII. Furthermore, these enzymes act as fibrinolytic agents, which also cleave fibrinogen. Leukocyte elastase attacks the platelet glycoproteins Ib (the receptor for von Willebrand factor) and V (a thrombin substrate) and at the same time exposes the platelet fibrinogen receptor. Platelet-activating factor of leukocyte origin may induce platelet aggregation and is a powerful potentiator of other inducers of platelet activity. The last-mentioned property has also been reported for leukotrienes. Activated leukocytes and their products play an important role in the pathogenesis of important disorders of the hemostatic system, with disseminated intravascular coagulation as the most prominent example.

Isocitrate dehydrogenase 1/2 mutant (IDHmt) astrocytoma is considered a T cell-deprived tumor, yet little is known regarding the phenotypes underlying T cell exclusion. Using bulk, single nucleus and spatial RNA and protein profiling, we demonstrate that a distinct spatial organization underlies T cell confinement to the perivascular space (T cell cuff) in IDHmt astrocytoma. T cell cuffs are uniquely characterized by a high abundance of gemistocytic tumor cells (GTC) in the surrounding stroma.

Granulocyte colony stimulating factor (G-CSF) has been shown in mammalia to activate a series of signal transduction systems and exert various biological effects, such as controlling the differentiation, proliferation, and survival of granulocytes, promoting the movement of hematopoietic stem cells from the bone marrow to the bloodstream, and triggering the development of T cells, dendritic cells, and immune tolerance in transplants. In this study, the mRNA of flounder G-CSF (PoG-CSF) and its receptor (PoGCSFR) were detected and widely expressed in all examined tissues with the highest expression in peritoneal cells. G-CSF and GCSFR cells were observed to be abundantly distributed in the leukocytes from the peritoneal cavity, followed by head kidney.

At this stage in the COVID-19 pandemic, most infections are "breakthrough" infections that occur in individuals with prior severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) exposure. To refine long-term vaccine strategies against emerging variants, we examined both innate and adaptive immunity in breakthrough infections. We performed single-cell transcriptomic, proteomic, and functional profiling of primary and breakthrough infections to compare immune responses from unvaccinated and vaccinated individuals during the SARS-CoV-2 Delta wave.

Cancer immunotherapies rely on CD8 cytolytic T lymphocytes (CTLs) in recognition and eradication of tumor cells via antigens presented on major histocompatibility complex class I (MHC-I) molecules. However, we observe MHC-I deficiency in human and murine urologic tumors, posing daunting challenges for successful immunotherapy. We herein report an unprecedented nanosonosensitizer of one-dimensional bamboo-like multisegmented manganese dioxide@manganese-bismuth vanadate (BMMBV) to boost multiple branches of immune responses targeting MHC-I-deficient tumors.
