AI Article Synopsis

  • Previous research indicated that high levels of Forkhead box D4 (FOXD4) in colorectal cancer lead to poor patient outcomes, but further diagnostic and mechanistic studies were lacking.
  • This study analyzed FOXD4 expression in colon adenocarcinoma patients using data from The Cancer Genome Atlas and created a nomogram for predicting patient survival based on FOXD4 levels.
  • Findings suggest that FOXD4 could serve as a diagnostic biomarker for colorectal cancer, with low expression linked to better clinical outcomes and numerous genes associated with FOXD4 involved in various cancer-related biological processes.

Article Abstract

Previous studies found that Forkhead box D4 () overexpressed in human colorectal cancer had the worst prognosis. However, the diagnostic value and further mechanism have not been fully researched. Statistical examinations for expression colon adenocarcinoma (COAD) patients were obtained from The Cancer Genome Atlas (TCGA). Survival analysis was used to assess its prognostic value. Nomogram model was used for visual prediction of patient survival rate. The online functional enrichment analysis tool was used to evaluate the biological functions and pathways of FOXD4 and its co-expressed genes. Receiver operating characteristic curve analysis suggested that FOXD4 might be a diagnostic biomarker for COAD (<0.001, area under the curve [AUC]=0.728, 95% confidence interval [CI]=0.669-0.787). Low expression of was associated with a good clinical outcome (=0.001, HR=0.517, 95% CI=0.341-0.782). A total of 797 genes were correlated with and associated with cell proliferation, cell differentiation, nuclear matrix, Rap1 signaling pathway, RNA transport, and VEGF signaling pathway. In conclusion, expression of may be a diagnostic and prognostic biomarker in COAD.

Foot ulcers are amongst the most prevalent complications of diabetes, known for their delayed healing process. Recent research indicates that the transcription factor forkhead box M1 (FOXM1) plays a role in promoting diabetic ulcer repair. However, the precise mechanisms underlying FOXM1 functions in this context remain unclear.

Osteoarthritis (OA) is a common age-related disease that is correlated with a high number of senescent chondrocytes in joint tissues. Heterogeneous nuclear ribonucleoprotein D (HNRNPD) is an RNA-binding protein whose expression imbalance is associated with cell senescence, but the role of HNRNPD in the occurrence and development of OA has not been reported. In this study, HNRNPD was found to be associated with the chondrocyte senescence process.

Objective: To investigate the relationship between the expression levels of serum forkhead box protein M1(FOXM1) and insulin-like growth factor 2 (IGF2) mRNA in patients with acute respiratory distress syndrome (ARDS) condition and prognosis.

Methods: Ninety patients with ARDS admitted to our hospital were regarded as the ARDS group, according to the prognosis, they were grouped into death group ( = 64) and survival group ( = 126); the control group consisted of 190 healthy individuals.

Results: Compared with the control group, the level of serum FOXM1 mRNA in ARDS group was obviously lower, and the level of IGF2 mRNA was higher.

In this study, two high-content flavonoid derivatives [3-8 biapigenin (HM 104) and quercetin-3--β--galactopyranoside (HM 111)] were obtained through the bioactivity-guided isolation of antidiabetic compounds from flowers. HM 104 and HM 111 exhibited good glucose consumption in fatty acid-induced insulin-resistant HepG2 cells. Moreover, both active compounds enhanced glucose uptake by restoring the expression of key regulators of glucose metabolism, including insulin receptor substrate 1, phosphoinositide 3-kinase, protein kinase B, and glucose transporter type 4, and by mitigating the expression of forkhead box O1 and the factors involved in gluconeogenesis.

Systemic lupus erythematosus (SLE) is a chronic and systemic autoimmune disease characterized by dysregulation in both innate and adaptive immunity. Polarization of macrophages into M1/M2 macrophages affects the development of lupus. Exosomes-miRNA plays a crucial role in disease progression.
