SNPs in miRNA machinery genes may affect miRNA function by impacting their biogenesis. Here, we investigated the association between three SNPs in miRNA machinery genes ( rs1057035, rs10719 and rs11077) and bicuspid aortic valve (BAV). Three polymorphisms were analyzed in 177 BAV patients and 414 healthy subjects by using a TaqManSNP assay. The frequencies of rs11077 genotype were significantly different between BAV patients and controls (p = 0.022). On multivariate logistic regression analysis, the rs11077 C allele resulted a significant predictor of BAV (odds ratio = 0.65; CI: 0.42-0.98; p = 0.047). The rs11077 SNP was associated with a decreased BAV risk supporting the causative role of miRNAs in aortic valve development.

Background: We aimed to elucidate the potential correlation between single-nucleotide polymorphisms (SNPs) in miRNA machinery genes and colorectal cancer (CRC) risk in an Iranian cohort.

Methods: We conducted a robust case-control study involving 507 participants, which included 213 patients diagnosed with CRC and 294 healthy controls at Research Institute for Gastroenterology and Liver Diseases in Tehran Province, Iran in 2018. The study focused on genotyping four specific SNPs, (rs14035), (rs197412), (rs2740348), and (rs3742330), using advanced ARMS-PCR and Tetra-primer ARMS-PCR techniques.

Non-coding RNAs (ncRNAs) play crucial roles in gene expression regulation, translation, and disease development, including cancer. They are classified by size in short and long non-coding RNAs. This chapter focuses on the functional implications of adenosine-to-inosine (A-to-I) RNA editing in both short (e.

MicroRNAs (miRNAs) are small, yet profoundly influential, non-coding RNAs that base-pair with mRNAs to induce RNA silencing. Although the basic principles of miRNA biogenesis and function have been established, recent breakthroughs have yielded important new insights into the molecular mechanisms of miRNA biogenesis. In this Review, we discuss the metazoan miRNA biogenesis pathway step-by-step, focusing on the key biogenesis machinery, including the Drosha-DGCR8 complex (Microprocessor), exportin-5, Dicer and Argonaute.

microRNAs (miRNAs) and small interfering RNAs (siRNAs) are 21- to 22-nucleotide RNAs that guide Argonaute-class effectors to targets for repression. In this work, we uncover 5-aminolevulinic acid synthase 1 (ALAS1), the initiating enzyme for heme biosynthesis, as a general repressor of miRNA accumulation. Although heme is known to be a positive cofactor for the nuclear miRNA processing machinery, ALAS1-but not other heme biosynthesis enzymes-limits the assembly and activity of Argonaute complexes under heme-replete conditions.

The excision of specific tRNA-derived small RNAs (tsRNAs), yRNA-derived small RNAs (ysRNAs) and ribosomal RNA-derived small RNAs (rsRNAs) is now well established. Several reports have suggested many of these fragments function much like traditional microRNAs (miRNAs). That said, whereas the expressions of the majority of appreciably expressed miRNAs in HCT116 colon cancer cells are significantly decreased in individual knockouts (KOs) of DROSHA, DGCR8, XPO5, and DICER, on average, only 3.
