Cell therapies are becoming increasingly widely used, and their production and cryopreservation should take place under tightly controlled GMP conditions, with minimal batch-to-batch variation. One potential source of variation is in the thawing of cryopreserved samples, typically carried out in water baths. This study looks at an alternative, dry thawing, to minimise variability in the thawing of a cryopreserved cell therapy, and compares the cellular outcome on thaw. Factors such as storage time, patient age, and gender are considered in terms of cryopreservation and thawing outcomes. Cryopreserved leukapheresis samples from 41 donors, frozen by the same protocol and stored for up to 17 years, have been thawed using automated, water-free equipment and by conventional wet thawing using a water bath. Post-thaw viability, assessed by both trypan blue and flow cytometry, showed no significant differences between the techniques. Similarly, there was no negative effect of the duration of frozen storage, donor age at sample collection or donor gender on post-thaw viability using either thawing method. The implication of these results is that the cryopreservation protocol chosen initially remains robust and appropriate for use with a wide range of donors. The positive response of the samples to water-free thawing offers potential benefits for clinical situations by removing the subjective element inherent in water bath thawing and eliminating possible contamination issues.

The application of cellulose nanofibers (CNF) as cryoprotectants in frozen foods has rarely been explored. In this study, the cryoprotective effect of CNF (2, 4 and 6 % w/w) on mechanically separated chicken meat (MSCM) surimi-like material was investigated, during frozen storage (5 and 60 days) under temperature fluctuation. Surimi-like without cryopreservation agents was more susceptible to protein oxidation due to ice recrystallization.

Study Question: Are live birth rates (LBRs) per woman following flexible progestin-primed ovarian stimulation (fPPOS) treatment non-inferior to LBRs per woman following the conventional GnRH-antagonist protocol in expected suboptimal responders undergoing freeze-all cycles in assisted reproduction treatment?
Summary Answer: In women expected to have a suboptimal response, the 12-month likelihood of live birth with the fPPOS treatment did not achieve the non-inferiority criteria when compared to the standard GnRH antagonist protocol for IVF/ICSI treatment with a freeze-all strategy.
What Is Known Already: The standard PPOS protocol is effective for ovarian stimulation, where medroxyprogesterone acetate (MPA) is conventionally administered in the early follicular phase for ovulatory suppression. Recent retrospective cohort studies on donor cycles have shown the potential to prevent premature ovulation and maintain oocyte yields by delaying the administration of MPA until the midcycle (referred to as fPPOS), similar to GnRH antagonist injections.

The aim of this work is to study the effect of adding hydroxy derivatives of chalcones to the basic cryomedium on the ability of sterlet sperm to utilize superoxide and hydrogen peroxide, the intensity of lipid peroxidation of male fish germ cells, and their viability both before cryopreservation and after 3 days of freezing at liquid nitrogen temperature. The ability of phenolic derivatives of chalcones to increase the superoxide dismutase and catalase activities of sterlet sperm and to reduce the intensity of lipid peroxidation has been established. The antioxidant activity of the derivatives exceeds the effect of Trolox, which inhibits the functioning of the enzyme component of the antioxidant protection of fish sperm and promotes lipid peroxidation of fish sperm before cryopreservation.

Efficient cryopreservation of stem cells is crucial to fabricating off-the-shelf cell products for tissue engineering and regeneration medicine. However, it remains challenging due to utilization of toxic cryoprotectants for reducing ice-related cryodamages to stem cells during freeze-thaw cycle, stringent post-thaw washing process, and further integration of stem cells with scaffolds to form tissue engineering constructs for downstream applications. Herein, a novel cryopreservation platform of stem cells based on an antifreezing polyvinylpyrrolidone/gellan gum/gelatin (PGG) scaffold together is reported with an L-proline assisted cell pre-dehydration strategy.
