A series of dipeptide analogues of angiotensinogen have been prepared and evaluated for their ability to inhibit the aspartic proteinase renin. The compounds were derived from the renin substrate by replacing the scissile amide bond with a transition-state mimic and by incorporating bioisosteric replacements for the Val-10 amide bond. Analogue 21a exhibited an IC50 of 7.6 nM against purified human renin, showed high specificity for this enzyme, and produced a hypotensive response in anesthetized, salt-depleted cynomolgus monkeys.

Diethylene glycol dimethacrylate (DEG)-crosslinked polystyrene (PS) resin offers a promising alternative to traditional divinyl benzene (DVB)-PS resin for solid-phase peptide synthesis (SPPS), particularly for challenging sequences with hydrophobic or bulky amino acids. The ultrashort peptide -fluorenylmethoxycarbonyl-phenylalanyl-phenylalanine (FmocFF) has been largely investigated due to its ability to self-assemble into fibrils (100 nm-μm scale) that can form a sample-spanning gel network. The initiation of the gelation process requires either a solvent switch (water added to dimethyl sulfoxide) or a pH-switch (alkaline to neutral) protocol, both of which ensure the solubility of the peptide as a necessary step preceding gelation. While the respective gel phases are well understood in structural and material characteristics terms the pregelation conditions are known to a lesser extent.

Imidazole dipeptides (IDPs) are found in the skeletal muscles and brains of various animals, and they exhibit unique functions like antioxidant and antiaging properties. Despite their importance, the metabolic mechanisms and physiological roles of IDPs remain unclear. Herein, we propose a method for the simultaneous analysis of IDPs, their constituent amino acids, and taurine, which are difficult to separate using conventional labeling reagents or columns, using liquid chromatography-single quadrupole mass spectrometry with PBr column and our highly sensitive labeling reagent, 1-fluoro-2,4-dinitrophenyl-5-l-valine-,-dimethylethylenediamineamide (l-FDVDA).
